Abstract
Stroke iskemik merupakan penyakit yang terjadi bila pembuluh darah yang memasok darah ke otak tersumbat oleh gumpalan darah sehingga sel-sel di daerah injury tidak menerima oksigen dan glukosa yang dibutuhkan. Antioksidan merupakan senyawa atau molekul yang dapat mencegah terjadinya proses oksidasi yang disebabkan oleh radikal bebas. Antosianin pada ubi jalar ungu berfungsi sebagai antioksidan dan penangkap radikal bebas, sehingga berperan untuk mencegah terjadinya stroke. Penelitian ini bertujuan untuk mengetahui pengaruh terapi antosianin ubi jalar ungu (Ipomoea batatas L) terhadap ekspresi Brain Derived Neurotrophic Factor (BDNF) dan ekspresi Tropomyosin receptor kinase B (TrkB) pada cerebrum tikus (Rattus norvegicus) model stroke iskemik. Hewan coba yang digunakan yaitu tikus putih (Rattus novergicus) umur 8-12 minggu dan berat rata-rata 200 gram. Tikus dibagi menjadi 5 kelompok perlakuan, yaitu kelompok kontrol negatif, stroke iskemik 24 jam, stroke iskemik 72 jam, terapi stroke iskemik 24 jam dan 72 jam dengan dosis 4 mg/kg berat badan. Pengamatan ekspresi BDNF dan TrkB dilihat menggunakan metode imunohistokimia. Analisa data ekspresi BDNF dan TrkB menggunakan analisa ragam ANOVA dan uji lanjutan Tukey untuk melihat ada tidaknya perbedaan nyata. Hasil penelitian menunjukkan pemberian terapi 72 jam antosianin dari ubi jalar ungu secara signifikan (p<0,05) dapat meningkatkan area ekspresi Brain derived neurotrophic factor (BDNF) sebesar 73, 44 % dan meningkatkan area ekspresi Tropomyosin receptor kinase B (TrkB) sebesar 75,30%. Kesimpulan dari penelitian ini menunjukkan antosianin dari ubi jalar ungu dapat digunakan sebagai terapi alternatif pada tikus model stroke iskemik.
English Abstract
Ischemic stroke is a disease that occurs when blood vessels that supply blood to the brain are clogged by blood clots so that cells in the injury region do not receive the required oxygen and glucose. Antioxidants are compounds or molecules that can prevent the oxidation process caused by free radicals. The anthocyanin compound in purple sweet potato serves as an antioxidant and free radical catcher, thus contributing to prevent of stroke. The aim of this study was to determine the effect of purple sweet potato anthocyanin therapy (Ipomoea batatas L) on expression of Brain Derived Neurotrophic Factor (BDNF) and expression of Tropomyosin receptor kinase B (TrkB) on rat cerebrum (Rattus norvegicus) ischemic stroke model. The experimental animals used were white rats (Rattus novergicus) aged 8-12 weeks and average weight of 200 grams. The rats were divided into 5 groups, negative control group, ischemic stroke of 24 h, ischemic stroke of 72 h, ischemic stroke therapy 24 h and 72 h with a dose of 4 mg/kg. Expressions of BDNF and TrkB were measured by immunohistochemistry method. Data of BDNF and TrkB expressions area were analyzed by using anova test followed by honestly significant difference test (HSD). The results showed that 72 h anthocyanin therapy from purple sweet potato significantly (p <0.05) increased the expression area of Brain derived neutrophic factor (BDNF) up to 73,44% and increased expression area of Tropomyosin receptor kinase B (TrkB) up to 75,30%. In conclusions : anthocyanin from purple sweet potato can be used as an alternative therapy in ischemic stroke based on BDNF and TrkB expressions.
