Abstract
Peningkatan produktifitas perlu dilakukan pada sapi potong di Indonesia salah satunya sapi Madura (bos indicus) dan Simmental (bos taurus), dimana ras sapi ini mendominasi penyediaan daging sapi di Indonesia.Kariotypememilikiperananpenting dalamseleksi yaknidalammengamatisifatketurunandenganmenentukankelainan yang terdapatpadaanatomi, morfologidanfisiologi yang disebabkanolehkelainankromosom. Tujuan dari penelitian ini yaituuntukmengetahuistrukturdanjumlahkromosomsapipejantanIB Madura dan Simmental sebagaiidentifikasisecaragenetik. Jenis sapi yang digunakan diperoleh dari Balai Besar Inseminasi Buatan Singosari (BBIB Singosari) yakni sapi Madura dan sapi Simmental. Sampel yang diambil berupa whole blood. Kemudian sampel dipreparasi melalui tahap kultur, harvest, preparasi slide dan pengamatan kromosom dibawah mikroskop cahaya. HasilpenelitianmenunjukkansapiMaduramemiliki 30 pasangkromosomdengansentromerpada autosomal kromosomberadapadaposisiakrosentrik.SekskromosomX berada padasubmetasentrikdan Y memilikisentromerpadaposisi Akrosentrik.SapiSimmentalmenunjukkanhasil yang sama untuk letak sentromer autosomal namun menunjukkan hasil berbeda dengan letak sentromer seks kromosom. SapiSimmentalmemilikikromosomdenganjumlah 30 pasang.Letaksentromer autosomal kromosomterletak di posisiakrosentrik.Sentromeruntuk sekskromosom X berada di posisisubmetasentrikdankromosom Y di submetasentrik. Penelitian ini menunjukkan kondisi kromosom pada sapi pejantan yang digunakan sebagai inseminasi buatan sapi Madura dan sapi Simmental memiliki kromosom yang normal.
English Abstract
Increase in productivity need to be done for beef cattle in Indonesia especially for Simmental(bos indicus) and Simmental (bos taurus) breed,.Karyotype has an important role in cytogenetic studies for observing offspring character by determining the defect in anatomy structure and physiology caused by chromosome abnormality. The purpose of this study is to identify genetic condition of artificial insemination bull specifically breed Madura and Simmental based on structure and total number of chromosome. Cattles that was used in this experimentation were from BalaiBesarInseminasiBuatanSingosari (BBIB Singosari) which consist of Simmental and Madura breed. Whole blood was collected as the samples. Preparation of the samples consist of culture, harvest, slide preparation, and chromosome observation under light microscope. Result shown that Madura cattle had 30 pair of chromosomes. Autosomal chromosomes of Madura cattle had centromere in acrocentric position while sex chromosome X and Y had centromere in sub metacentric and acrocentric position respectively. Simmental cattle shown the same result for centromere position of autosomal chromosome but shown different result for centromere position of sex chromosome. Simmental cattle had 30 pair of chromosomes. Centromere for autosomal chromosomes were on acrocentric position. Centromere for sex chromosome X was on sub metacentric position and chromosome Y on the same position which was sub metacentric position. This study shown that sample Artificial insemination bull from Simmental and Madura breed had normal chromosomes.
