Abstract

Glomerulonefritis akut salah satu penyakit disebabkan kompleks imun ditandai inflamasi glomerulus. Aktivasi komplemen dari kompleks imun menyebabkan peningkatan radikal bebas yang menyebabkan stres oksidatif. MDA merupakan hasil peroksidasi lipid akibat radikal bebas. Limpa merupakan salah satu organ pertahanan tubuh yang peran dalam proses penyembuhan pada sistemik inflamasi pada GNA. Salah satu terapi GNA adalah antiinflamasi dan antioksidan. Daun kemangi memiliki kandungan flavonoid, eugenol dan tanin sebagai antiinflamasi dan antioksidan. Penelitian bertujuan mengetahui kadar MDA serta perubahan histopatologi limpa mencit model GNA hasil induksi streptokinase yang diterapi ekstrak daun kemangi. Penelitian eksperimen ini menggunakan Rancangan Acak Lengkap dengan post test control design only. Menggunakan mencit jantan 6-8 minggu, berat 20-25 gram, dibagi 5 kelompok perlakuan. Kontrol negatif, kontrol positif diinduksi streptokinase dosis 2500 IU serta kelompok terapi dosis 400, 800 dan 1200 mg/kg BB. Pemeriksaan kadar MDA ginjal dengan Spektofotometer dan histopatologi limpa dengan pewarnaan HE. Kadar MDA dianalisa secara kuantitatif dengan One Way ANOVA (α=0,05) serta analisa deskriptif untuk histopatologi limpa. Hasil penelitian menunjukkan ekstrak etanol daun kemangi dosis 400 mg/kg BB dapat menurunkan kadar MDA ginjal secara signifikan (p<0,05) dan dapat menurunkan infiltrasi giant cell serta menurunkan tingkat kerusakan organ limpa. Kesimpulan dari penelitian ini pemberian ektrak etanol daun kemangi dapat digunakan sebagai terapi GNA.

English Abstract

Acute glomerulonephritis is one of diseases caused immune complex marked by glomerular inflammation. Complement activation of the immune complex causes an increase in free radicals that cause oxidative stress. MDA is the result of lipid peroxidation due to free radicals. The spleen is one of the body's defense organs that plays a role in the process of healing the systemic inflammation in GNA. One of the GNA therapies is anti-inflammatory and antioxidant. Basil leaves contain flavonoids, eugenol and tannins as anti-inflammatory and antioxidants. The aim of this research was to know the level of MDA and histopathologic changes of spleen mice of GNA model of induction of streptokinase treated by basil leaf extract. This experimental study used a Completely Randomized Design with post test control design only. Using male mice 6-8 weeks, weight 20-25 grams, divided by 5 treatment groups. Negative control, positive control induced streptokinase dose 2500 IU and dose therapy group 400, 800 and 1200 mg / kg BW. Examination of MDA levels of the kidney with Spectrophotometer and histopathology of the spleen by HE staining. MDA levels were analyzed quantitatively with One Way ANOVA (α = 0.05) and descriptive analysis for histopathology of the spleen. The results showed that basil leaves ethanol extract of 400 mg / kg BW could significantly decrease the MDA level of kidneys (p <0.05) and can decrease giant cell infiltration and decrease lymph organ damage rate. The conclusion of this research giving the extract of basil leaf ethanol can be used as GNA therapy.
