Abstract
Rheumatoid arthritis (RA) adalah peradangan pada bagian persendian. Penelitian ini menggunakan ekstrak daun tempuyung (Sonchus arvensis) sebagai terapi pada hewan tikus (Rattus novergicus) model RA yang diinduksi oleh Complete Freund’s Adjuvant (CFA). Kandungan flavonoid pada ektrak daun tempuyung dapat menghambat radikal bebas dan menghambat pengeluaran sitokin proinflamatori. Tujuan penelitian ini untuk mengetahui ekspresi interferon-gamma (IFN- γ) dan mengetahui aktivitas protease hati pada hewan tikus model RA setelah diberi terapi ekstrak daun tempuyung (Sonchus arvensis). Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan lima kelompok perlakuan, yaitu kelompok kontrol negatif (A), kelompok kontrol positif (B) dan kelompok terapi ekstrak daun tempuyung yang terdiri atas pemberian dosis yang berbeda yaitu 50 mg/kg BB (C), 100 mg/kg BB (D) dan 150 mg/kg BB (E). Tikus model RA disiapkan dengan cara pemberian induksi CFA sebanyak dua kali, 0,1 ml/ekor pada hari ke-1 di coccygea dan 0,05 ml/ekor pada hari ke-8 di metatarsophalangeal. Setiap kelompok perlakuan menunjukkan perbedaan yang signifikan (p<0,05). Perlakuan pemberian CFA menyebabkan inflamasi dan meningkatkan ekspresi IFN-γ sebesar 184,6 %. Pemberian ekstrak daun tempuyung 200 mg/Kg BB menunjukkan penurunan ekspresi IFN-γ sebasar 36,8 % dan penurunan aktivitas protease hati sebesar 6,8 % pada tikus arthritis. Jadi ekstrak daun tempuyung (Sonchus arvensis L.) berpotensi untuk dijadikan sebagai alternatif pengobatan artritis.
English Abstract
Rheumatoid arthritis (RA) is an inflammation of the joints. This study used tempuyung leaves extract (Sonchus arvensis) as a therapy in rats (Rattus novergicus) which induced with Complete Freunds Adjuvant (CFA). Flavonoids in sonchus leaves extract inhibit free radicals and inhibit proinflammatory cytokines secretion. The purpose of this experiment was to determine the expression of interferon-gamma (IFN-γ) and to know the protease activity of rat liver in animal models of RA after treatment sonchus leaves extract. This study used a completely randomized design (CRD) with five treatment groups, the negative control group (A), positive control group (B) and groups with therapy of sonchus leaf extract given with dose of: 50 mg / kg body weight (C) , 100 mg / kg (D) and 150 mg / kg (E). Rat models of RA were prepared by injection of CFA twice, 0.1 ml / rat on day 1 in coccygea and 0.05 ml / rat on day 8 in metatarsophalangeal. Statistical analysis showed that every treatment group showed a significant difference( p < 0.05 ). CFA treatment caused inflammation and increased the expression of IFN - γ to be 184.6 %. Sonchus leaf extract therapy dose of 200 mg / kg bw showed decreasing expression of IFN - γ to be 36.8 % and a decreasing of protease activity to 6.8% in rat arthritis model. Thus can be said that sonchus leaf extract (Sonchus arvensis L.) had a potential for arthritis treatment .
