Abstract

Kanker mammae adalah tumor ganas yang menyerang organ mammae. Pembuatan hewan model kanker mammae dapat menggunakan zat karsinogenik 7,12-Dimethylbenz(ɑ)antrasene (DMBA). Induksi DMBA pada mammae menyebabkan kerusakan DNA, akumulasi ROS dan kenaikan VEGF sehingga dimungkinkan dapat mempengaruhi kadar VEGF dan ekspresi IL-2 pada uterus tikus. Kombinasi kurkumin dan vitamin E merupakan antioksidan yang dapat digunakan sebagai terapi kanker. Penelitian ini bertujuan untuk mengetahui pengaruh terapi kombinasi kurkumin dan vitamin E terhadap kadar VEGF dan ekspresi IL-2 pada uterus tikus hasil induksi DMBA. Penelitian ini menggunakan tikus (Rattus norvegicus) strain sprague dawley betina dengan umur 10-12 minggu dan berat badan rata-rata 150-200 gram. Pembuatan hewan model kanker mammae dilakukan dengan induksi DMBA dosis 10 mg/kg BB secara injeksi subcutan pada mammae sebanyak 1ml dan induksi estrogen dengan dosis 20.000 IU/kg BB secara intramuscular. Penelitian ini dibagi menjadi 5 kelompok yaitu Kontrol negatif (KN), kontrol positif (KP), Terapi 1 (kurkumin 48mg/kg BB + vitamin E 300 IU/ekor), Terapi 2 (kurkumin 72mg/kg BB + vitamin E 200 IU/ekor), Terapi 3 (kurkumin 108mg/kg BB + Vitamin E 100 IU/ekor). Parameter yang diamati adalah Kadar VEGF dengan metode ELISA dan ekspresi IL-2 dengan metode imunohistokimia. Hasil penelitian menunjukkan bahwa kombinasi terapi kurkumin dan vitamin E mampu menurunkan kadar VEGF dan ekspresi IL- 2 secara signifikan (p<0,05) dengan dosis terapi 2 (kurkumin 72mg/kg BB + vitamin E 200 IU/ekor) yang paling efektif dalam menurunkan kadar VEGF sebesar (84,40 ± 6,06 ng/L) dan menurunkan ekspresi IL-2 sebesar (6,21 ± 0,67%). Kesimpulan dari penelitian ini adalah kombinasi kurkumin dan vitamin E dapat digunakan sebagai terapi kanker mammae.

English Abstract

Mammary cancer is the malignant cancer which attack on mammary organs. The production of animal mammary cancer model used carcinogenic substances 7,12- Dimethylbenz (a) antrasene (DMBA). The induction of DMBA on mammary can caused DNA damage, ROS acummulated and the tension of VEGF so it can possibly influence the VEGF level and the expression of IL-2 on the rat uterine. The combination of curcumin and vitamin E are the antioxidants those can be used for cancer therapy. The purpose of the research was to investigate the effect of curcumin combination therapy and vitamin E toward VEGF level and expression of IL-2 on the rat uterine the result of DMBA induction. This research used rats (Rattus norvegicus) strain sprague dawley female which the age of 10- 12 weeks old and the weight of 150-200 gram. The production of animal mammary cancer model conducted by giving induction of DMBA doses 10 mg/kg BW by subcutaneusly injection toward mammae and followed by intramuscularly injection estrogen doses 20.000 IU/kg BW. This research divided into 5 groups, they were negative control (KN), postive control (KP), therapy 1 (curcumin 48 mg/kg of BW + Vitamin E 300 IU/rat), therapy 2 (curcumin 72mg/kg of BW + vitamin E 200 IU/rat), therapy 3 (curcumin 108mg/kg BW + vitamin E 100 IU/rat). The parameter that being observed were VEGF level with the ELISA method and expression of IL-2 with the immunohistochemistry method. The result of the research showed that the combination of curcumin therapy and vitamin E were able to reduce VEGF level and expression of IL-2 with the dosis of therapy 2 significantly (p<0,05) (curcumin 72mg/kg BB of BW + vitamin E 200 IU/rat). It was the most effective in reducing VEGF level (84,40 ± 6,06 ng/L) and the expression of IL-2 (6,21 ± 0,67%). The conclusion of the research were the combination of curcumin and vitamin E were able to use as the mammary cancer therapy.
