Abstract
Penyakit kardiovaskuler menempati urutan pertama penyebab kematian di dunia. penyakit kardiovaskuler banyak disebabkan oleh hiperkolesterolemia. Upaya mempertahankan tingkat kolesterol dalam tubuh yaitu dengan kosumsi yoghurt susu kambing yang mengandung bakteri asam laktat (BAL), dan biopeptida aktif, yang berfungsi sebagai antioksidan dan antiinflamasi. Mengetahui potensi pemberian yoghurt susu kambing dalam mempertahankan gambaran histopatologi dan ekspresi Inducible Nitric Oxide Synthase (iNOS) pada jantung tikus (Rattus norvegicus) yang diberi diet hiperkolesterol. Penelitian ini menggunakan Rattus norvegicus strain Wistar jantan yang dibagi menjadi 5 kelompok perlakuan yaitu kelompok kontrol, kelompok control diet hiperkolesterolemia, kelompok dosis yoghurt 300 mg/kg BB, 600mg/kg BB, dan 900 mg/kg BB. Diet hiperkolesterolemia dibuat dari campuran asam kolat, minyak babi, dan kuning telur puyuh rebus. Pencegahan hiperkolesterolemia dilakukan dengan pemberian yoghurt susu kambing selama 28 hari dan di lanjutkan dengan pemberian yoghurt susu kambing dan diet hiperkolesterolemia selama 14 hari. Analisa data secara kuantitatif dengan penghitungan presentase area ekpresi iNOS yang dianalisa menggunakan analysis of variance (ANOVA) dilanjutkan dengan uji Beda Nyata Jujur (Tukey) dengan α = 5%. Pengamatan histopatologi jaringan jantung disajikan dan dianalisa secara deskriptif. Hasil pada penelitian ini menunjukkan perbedaan nyata (p<0,05) pada ekspresi iNOS. Pencegahan hiperkolesterolimia melalui pemberian yoghurt susu kambing dengan dosis 900 mg/kg BB mampu menekan peningkatan ekpresi iNOS sebesar 48, 72%, serta mampu mempertahankan histopatologi jantung tikus kondisi hiperkolesterolimia. Kesimpulan dari penelitian ini, yoghurt susu kambing dapat dijadikan salah satu alternatif pencegahan hiperkolesterolimia.
English Abstract
Cardiovascular disease is the first ranks cause of death in the world. Cardiovascular diseases are caused by hypercholesterolemia. The body maintaining the level of cholesterol by consume goats milk yogurt containing lactic acid bacteria (BAL), and active biopeptide, functioned as an antioxidant and anti-inflammatory. This research aim to potential of goat milk yoghurt in maintaining expression of Inducible Nitric Oxide Synthase (iNOS) and histopathology in heart rat (Rattus norvegicus) were given hypercholesterolemia diet. This study used male Wistar strain Rattus norvegicus were divided into 5 groups: control group, hypercholesterolemia, hypercholesterolemia prevention with goat milk yogurt dose of 300 mg/kg, 600 mg/kg, and 900 mg/kg. Hypercholesterolemic diet were made by mixturing of cholic acid, lard, and poached quail egg yolk. Prevention treatment of hypercholesterolemia conducted by consuming goat milk yogurt for 28 days and followed by administered of goats milk yoghurt and hypercholesterolemic diet for 14 days. Quantitative analysis data by calculating the percentage of iNOS expression area were analyzed using ANOVA and followed by Tukey test with α = 5%. Histopathological observation of heart tissue are presented and analyzed descriptively. The results in this study showed significant differences (p <0.05) in the expression of iNOS. Preventive dose of 900 mg/kg was able to suppress increasing of iNOS expression by 48.72%, and it was able to maintains histopathology of rat heart. The conclusion of this study, goat milk yogurt could be used as an alternative prevention of hiperkolesterolimia.
