Abstract
Penyakit infeksius sering menginfeksi ayam, oleh karena itu dibutuhkan tindakan pencegahan, salah satunya menggunakan imunostimulator. Buah lerak (Sapindus rarak) memiliki kandungan aktif berupa saponin, alkaloid, steroid, triterpen dan flavonoid yang bersifat sebagai imunostimulan. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak metanol buah lerak sebagai imunostimulator pada ayam broiler strain Cobb yang diinfeksi Salmonella enteritidis (S.enteritidis). Penelitian ini merupakan penelitian eksperimental murni menggunakan metode Rancangan Acak Lengkap (RAL), dengan post test control designs only dan dibagi menjadi 5 kelompok perlakuan. Kelompok 1 adalah kontrol negatif (ayam sehat), kelompok 2 adalah kontrol positif (diinfeksi S.enteritidis), dan 3 kelompok lain yang diberikan ekstrak metanol buah lerak selama 14 hari dengan dosis yang berbeda, yaitu 5 mg/kg BB, 10 mg/kg BB, 15 mg/kg BB secara per oral, kemudian diinfeksi dengan dosis infeksi S.enteritidis 0,2.10⁸ CFU/ml. Parameter yang diukur yaitu nilai kadar MDA, profil limfosit dan histopatologi ileum. Nilai kadar MDA diukur menggunakan metode thiobarbituric acid (TBA), sedangkan histopatologi ileum diwarnai dengan HE. Data kadar MDA dan limfosit ileum dianalisis secara statistik menggunakan one way ANOVA, dengan α = 0,05. Histopatologi ileum dianalisis secara deskriptif. Hasil analisis statistik menunjukkan perlakuan dosis 5 mg/kg BB mampu menurunkan kadar MDA sebesar 449,00 μg/mL dan meningkatkan jumlah limfosit ileum, serta mencegah kerusakan vili, nekrosis dan edema pada ileum. Kesimpulan dari penelitian ini adalah ekstrak metanol buah lerak dapat berfungsi sebagai imunostimulator.
English Abstract
Chickens is relatively prone of infectious disease, therefore it needs prevention. One of the effort is administering immunostimulant. Lerak fruit (Sapindus rarak ) contains an active compound called saponin, alkaloide, steroid, flavonoid, triterpen that potentially functioning as an immunostimulant. The aim of this study was to identify the effect of lerak fruit methanol extract as immunostimulant toward the immune system of Cobb broiler strain that infected by Salmonella enteritidis (S. enteritidis). This research was an experimental study using Completely Randomized Design (CRD) with post test control design only and chikens are divided into 5 groups. Group 1 are negative control (healthy chikens), group 2 are positive control (infected by S.enteritidis), and 3 other group are chikens that given with lerak fruit methanol extract for 14 days with different doses 5 mg/kg BW, 10 mg/kg BW and 15 mg/kg BW oraly, then they were infected by S.enteritidis 0,2.10⁸ CFU/ml. The observed variables included the value of MDA levels, lymphocyte profile and histophatology of ileum. The value of MDA levels measured using thiobarbituric acids (TBA) methods, while histophatology of ileum was HE stained. Futhermore, MDA levels and lymphocyte ileum was calculated statistically using one-way ANOVA with α = 0,05. Histophatology of ileum analyzed in descriptive. The result of statistic analysis showed that a dose of 5 mg/kg BW could decreased MDA levels as much as 449,00 μg/mL and increased the number of lymphocytes and prevent ileum’s vili damage, necrosis and edema. Finally, this research concluded that lerak fruit methanol extract could be used as an immunostimulant.
