Abstract

Polyvinyl Chloride (PVC) merupakan jenis plastik yang banyak digunakan untuk pembuatan pipa air. Salah satu logam berat yang menjadi bahan pembuatan pipa adalah timbal. Air minum yang disalurkan melewati pipa yang berlapis Pb akan menyebabkan tingginya kandungan Pb yang terlarut dalam air. Keberadaan plumbum di dalam tubuh dapat menyebabkan berbagai gangguan kesehatan dan kerusakan sel, organ, atau jaringan terutama pada lambung. Pipa air yang digunakan adalah pipa “A”, “B” dan pipa “C”. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan pipa polyvinyl chloride (PVC) yang terpapar Plumbum (Pb) dalam air berdasarkan gambaran histopatologi lambung dan aktivitas protease organ lambung pada tikus putih (Rattus norvegicus). Penelitian ini dibagi dalam 5 kelompok perlakuan, yaitu kontrol negatif, kontrol positif dengan pemberian Pb 5 mg/ekor/hari selama 45 hari, kelompok 3 diberikan air yang sudah direndam selama 60 hari di pipa “A”, kelompok 4 diberikan air yang sudah direndam selama 60 hari di pipa “B”, dan kelompok 5 diberikan air yang sudah direndam selama 60 hari dipipa”C”. Aktivitas protease diukur dengan metode Walter kemudian dianalisa secara kuantitatif menggunakan uji one way ANOVA pada progam SPSS. Gambaran histopatologi lambung diwarnai dengan pewarnaan Hematoxylin Eosin (HE) kemudian dianalisa secara kualitatif deskriptif dengan melakukan pengamatan menggunakan mikroskop cahaya BX51. Hasil penelitian membuktikan bahwa diantara perlakuan penggunaan pipa PVC menunjukkan perbedaan yang sangat nyata (P<0,01) terhadap peningkatan protease dan gambaran histopatologi lambung. Penggunaan pipa PVC tipe “A” merupakan pipa terbaik yang tidak mempengaruhi aktivitas protease dan gambaran histopatologi lambung.

English Abstract

Polyvinyl Chloride (PVC) is a type of plastic that is mostly used for a manufacture of water pipes. One of heavy metals that utilized in pipe-making materials is Plumbum (Pb). Drinking water that supplied through the pipes which is contaminated by Plumbum (Pb) will cause the high-rated of Pb dissolved in the water. The existence of Plumbum (Pb) in the body can cause various health problems and damage cells, organs, or tissue, especially in the stomach. The water pipes that were employed in this study were pipe “A”, pipe “B”, and pipe “C”. This study is aimed at discovering the influence of the use of polyvinyl chloride (PVC) which contaminated by Plumbum (Pb) in the water based on a picture of gastric histopathology and protease activity in the gastric organ of white rat (Rattus norvegicus). This study was divided into 5 groups of treatments, namely negative control, positive control with the provision of 5 mg Pb /a rat/ day for 45 days, group 3 was supplied with the water that has been soaked in pipe “A” for 60 days, group 4 was supplied with the water that has been soaked in pipe “B” for 60 days, and group 5 was supplied with the water that has been soaked in pipe “C” for 60 days. Protease activity was measured by the Walter method, and then analyzed quantitatively by using one way ANOVA test by SPSS. The picture of gastric histopathology is colored by a Hematoxylin Eosin (HE) coloration, then it is analyzed using descriptive qualitative by conducting an observation through the BX51 light microscope. The results of the study proved that the treatment using PVC pipes showed the prominent distinction (P<0.01) towards the increasing of protease in the picture of gastric histopathology. The use of PVC pipe type “A” is the best pipe which can maintain protease activity and the picture of gastric histopathology.
