Abstract
Osteoarthritis (OA) adalah penyakit sendi degeneratif progresif yang berlangsung secara perlahan diikuti oleh hilangnya kartilago pada artikular dan terkikisnya tulang subchondralis serta pembentukan osteofit. Pengobatan OA menggunakan AINS (Analgetik Anti Inflamasi Non-Steroid) masih mempunyai efek samping seperti iritasi mukosa lambung, iritasi glomerulus, pembekuan darah, dan hipertensi. Obat alami berupa cangkang telur dan lemak kambing yang berpotensi sebagai terapi alternatif karena memiliki kandungan glukosamin dan omega-3. Tujuan penelitian yaitu mengetahui pengaruh glukosamin cangkang telur dan omega-3 lemak kambing dalam menyembuhkan OA pada hewan model tikus putih (Rattus norvegicus) hasil induksi papain, berdasarkan penurunan jumlah neutrofil dan ketebalan kartilago. Penelitian bersifat eksperimental laboratorik dengan metode post test only control group. Hewan model yaitu tikus putih jantan, strain wistar, berat badan 150-250 gram, yang terdiri dari 5 kelompok yaitu kontrol negatif, kontrol positif (OA hasil induksi papain 4% sebanyak 0,2 ml secara intra-artikular sebayak 3 kali induksi), dan perlakuan dengan dosis tetap 27 mg glukosamin dan dosis bertahap omega-3 pada P1, PII, PIII yaitu 1,8 mg, 3,6 mg dan 5,4 mg. Parameter yang diukur adalah jumlah neutrofil dan ketebalan kartilago histopatologi artikular sendi. Data dianalisa secara kuantitatif dengan uji one way ANOVA dengan tingkat signifikan 0,05. Hasil penelitian menunjukkan adanya penurunan jumlah neutrofil dan peningkatan ketebalan kartilago pada dosis kombinasi 27 mg glukosamin + 5,4 mg omega-3. Kesimpulnnya bahwa glukosamin cangkang telur dan omega-3 lemak kambing dapat digunakan sebagai terapi OA.
English Abstract
Osteoarthritis (OA) is a progressive degenerative joint disease which happen slowly followed by the loss of cartilage in articular and also it can erode the subchodral bone and shape the osteophyte. Treatment that use NSAID (Non-Steroidal Anti-Inflamatory Drugs) for OA asa side effect: complications on gastrointestinal tracts, cardiovascular, nuisance of gland function, coagulate blood, and hypertension. Thus, we need to develop another alternative medicine that has low side effect. One of animal by products is eggshell and goat fat which is really useful for alternative therapy by products to contain glucosamine and omega-3. This research was aimed to find the effect of eggshell glucosamine and omega-3 of goat fat for Osteoarthritis treatment using Rattus norvegicusas the model then induced by papain to oversaw on the reduction of neutrophils and the thickness of cartilage. This research was experimental using post test only control group design method. Moreover, this research used male wistar rats weighing 150-250 gram, which divided into 5 group: negative control, positive control (OA induced by 0,2 ml 4% papain intra-articular, induced 3 times), and treatment group of therapy 27 mg glucosamine in all of the group and omega-3 graded, PI ,PII, PIII (1,8 mg, 3,6 mg, 5,4 mg omega-3 respectively). The parameters that were neutrophils and thethickness of histopathology cartilage articular. The quantitative data was analyzed using one way ANOVA test with significance level (α=0, 05). The result of this research showed that there was decrease of neutrophils and the thickness of cartilage on combination dosage of 27 mg glucosamine + 5,4 mg omega-3. The conclution of this study, eggshell glucosamine and the omega-3 of goat fat had a therapy on animal model of OA.
