Abstract

Kontaminasi air oleh logam berat dapat bersifat toksik dan mempengaruhi berbagai aspek biologi dan ekologi. Salah satu logam berat yang banyak ditemukan adalah Pb. Plumbum (Pb) terdapat pada pipa air sebagai zat aditif untuk mencegah korosi dan apabila masuk kedalam tubuh dapat menimbulkan kerusakan organ salah satunya hepar. Hepar dapat mengalami kerusakan apabila terpapar Pb secara terus menerus. Penelitian ini bertujuan untuk mengetahui pengaruh paparan Pb dalam air yang direndam pada tiga merek pipa PVC dengan kadar Pb pada masing-masing pipa : Pipa A : 0 mg/L; Pipa B: 0,05 mg/L; dan Pipa C: 0,08 mg/L. Penelitian ini menggunakan tikus (Rattus norvegicus) jantan yang dibagi dalam 5 kelompok. Kelompok 1 adalah kelompok kontrol negatif, kelompok 2 adalah kelompok kontrol positif yang diberikan paparan Pb dosis 5mg/ekor/hari, kelompok 3, 4, dan 5 adalah tikus yang dipapar Pb dari masingmasing air rendaman pipa A, B, dan C. Paparan Pb dilakukan selama 45 hari. Gambaran histopatologi hepar diamati dengan menggunakan mikroskop cahaya dan kadar SGOT serta SGPT darah tikus diukur dengan metode spektrofotometri. Hasil penelitian menunjukkan bahwa pengaruh paparan Pb dalam air yang direndam dalam pipa merek “A” tidak menyebabkan kerusakan histopatologi hepar dan peningkatan kadar SGOT serta SGPT. Adanya paparan Pb dalam air rendaman pipa PVC secara signifikan (p<0,05) mampu meningkatkan kerusakan histopatologi hepar dan meningkatkan kadar SGOT serta SGPT. Kesimpulan dari penelitian ini bahwa pipa merek “A” memberikan hasil terbaik yang ditandai dengan gambaran histologi hepar normal dan kadar SGOT serta SGPT terendah apabila dibandingkan dengan pipa merek “B” dan “C”.

English Abstract

The contamination by the heavy metal gains toxicity and can affect in various aspects of biology and ecology. One of the heavy metal that often found is Pb. Plumbum (Pb) can be found in water pipe as the additional substance to prevent the corrosion which if enter into the body can cause the organ damage, such as liver. Liver can get the damage when exposed to Pb continuously. This study aimed to determine the effect of Pb exposure to water which soaked in three brands of PVC pipe contains Pb level each one: Pipe A: 0 mg / L; Pipe B: 0.05 mg / L; and Pipe C: 0.08 mg / L. In this study a male rat (Rattus norvegicus) was used then they were divided into 5 groups. Group 1 was the negative control group, Group 2 was the positive control group that given Pb exposure with dose of 5mg / rat / day, Group 3, 4, and 5 the rat were exposed to Pb from each of the soaked water pipe A, B, and C. The Pb exposure were carried out in 45 days. Histopathology view of the liver was observed by light microscope, and the change of SGOT level and SGPT level in blood were measured by spectrophotometry method. The results showed the effect of Pb exposure on water from pipe “A” brand not cause the damage of liver histopathology and increase the level of SGOT and SGPT. The exposure of Pb on water significantly (p <0.05) could increase the damage of liver histopathology and increased the level of SGOT and SGPT. The conclusion from this study was the usage of pipe "A" brand gave the best results which marked with normal histologic view of liver and SGOT level and SGPT level more lower than pipe “B” and “C” brand.
