Abstract
Hiperkolesterolemia adalah gangguan metabolisme kolesterol yang disebabkan penigkatan kadar kolesterol dalam darah. Pemberian ekstrak air daun sukun diyakini dapat menjadi alternatif obat herbal untuk pengobatan hiperkolesterolemia untuk mengurangi efek pemberian obat sintesis. Penelitian ini bertujuan untuk mengetahui profil kadar LDL dan histopatologi aorta tikus hiperkolesterolemia dengan terapi ekstrak air daun sukun. Penelitian ini menggunakan hewan coba tikus (Rattus norvegicus) jantan yang dikelompokkan menjadi 5 kelompok yaitu kelompok kontrol, kelompok hiperkolesterolemia, kelompok hiperkolesterolemia dengan terapi ekstrak air daun sukun 500 mg/KgBB, 1000 mg/KgBB dan 2000 mg/KgBB. Peningkatan kadar LDL (Low Density Lipoprotein) diukur dengan metode spektofotometri dan analisa data kadar LDL (Low Density Lipoprotein) menggunakan one way ANOVA dengan uji lanjutan Tukey. Perubahan histopatologi pembuluh darah aorta secara deskriptif dilihat dari metode pewarnaan Hematoksilin Eosin (HE). Hasil penelitian menunjukkan bahwa terapi ekstrak air daun sukun secara nyata (p<0,05) mampu menurunkan kadar LDL pada tikus model hiperkolesterolemia. Dosis 2000mg/kgBB adalah dosis optimal dengan penurunan kadar LDL sebesar 78,28%. Histopatologis aorta tikus yang diterapi dengan ekstrak air daun sukun menunjukkan adanya perbaikan sel endotel pada aorta tikus mendekati sel endotel pada pembuluh darah aorta normal. Kesimpulan dari penelitian ini adalah bahwa ekstrak air daun sukun mempunyai potensi menurunkan kadar LDL dan memperbaiki histopatologis aorta pada tikus model hiperkolesterolemia.
English Abstract
Hypercholesterolemia is a cholesterol metabolism disorder caused by the excess of cholesterol in blood. Treatment with breadfruit leaf liquid extract was considered as alternative herbal medicine for hypercholesterolemia side effect of treatment with synthetic medicines. This research aimed to learn about profile of LDL and aortic histopathology on hypercholesterolemic rats after breadfruit leaf extract therapy. This research used rats (Rattus norvegicus) divided into five groups, control group, hypercholesterolemia group, and hypercholesterolemia group with breadfruit leaf extract therapy dose of 500 mg/Kg BW, 1000 mg/Kg BW, and 2000 mg/Kg BW, respectively. The level of LDL (Low Density Lipoprotein) were measured by spectrophotometry and data were analyzed using one-way ANOVA and Tukey Test. The alteration of aortic histopathology were confirmed by staining method with Hematoxylin Eosin (HE) and were analyzed descriptively. The result indicated that breadfruit leaf extract therapy significanly (p < 0.05) reduced LDL level on hypercholesterolemic rats. The optimal doses of theraphy was 2000 mg/Kg BW with reduction of 78,28%. The breadfruit leaf extract theraphy improved histopathology of aortic which showed normalized of aortic endothelial cells. In conclusion, the breadfruit leaf extract therapy have a potential to reduce LDL level and improve aortic histopathology of the hypercholesterolemic rats.
