Abstract
Inflammatory Bowel Disease (IBD) merupakan penyakit peradangan kronis yang terjadi pada saluran pencernaan. Terjadinya IBD disebabkan oleh efek samping penggunaan obat antiinflamasi non-steroid (NSAID) seperti indometasin. Ekstrak kerang pisau (Solen spp.) yang diketahui mengandung flavonoid berpotensi sebagai bahan terapi IBD. Tujuan penelitian ini adalah untuk mempelajari potensi terapi ekstrak kerang pisau (Solen spp.) terhadap penurunan ekspresi TNF-α dan gambaran histopatologi kolon tikus Inflammatory Bowel Disease (IBD). Tikus IBD diinduksi menggunakan indometasin dosis 15 mg/kg BB secara per oral. Penelitian ini menggunakan empat kelompok yaitu kelompok kontrol negatif, kelompok kontrol positif (IBD), kelompok terapi ekstrak kerang pisau dosis 100 mg/kg BB dan dosis 400 mg/kg BB. Penurunan ekspresi TNF-α diamati menggunakan metode imunohistokimia dan dilakukan analisa statistik ANOVA dilanjutkan uji BNJ (Beda Nyata Jujur) α = 0,05. Pengamatan gambaran histopatologi kolon diwarnai pewarnaan Hematoxylen-eosine (HE) diamati menggunakan mikroskop cahaya dan dilakukan analisis secara deskriptif. Hasil penelitian menunjukkan bahwa ekstrak kerang pisau (Solen spp.) memperbaiki histopatologi kolon tikus (Rattus norvegicus) Inflammatory Bowel Disease (IBD) melalui regenerasi epitel dan secara signifikan (p<0,05) menurunkan ekspresi TNF-α. Dosis terapi 400 mg/kg BB menurunkan ekspresi TNF-α tertinggi yaitu sebesar 55,41%. Kesimpulan penelitian ini adalah ekstrak kerang pisau (Solen spp.) dapat digunakan sebagai terapi alternatif Inflammatory Bowel Disease (IBD).
English Abstract
Inflammatory Bowel Disease (IBD) is a chronic inflammatory disease that occurs in the gastrointestinal tract. The case of IBD is caused by the use of nonsteroidal anti-inflammatory drugs (NSAIDs) side effects such as indomethacin. Solen spp. extracts which known containing flavonoid is potentially used as a therapy in IBD. The purpose of this research was to study the therapeutic of Solen spp. extracts in order to decrease the expression of TNF-α and colonic histopathology in rats of Inflammatory Bowel Disease (IBD). Rats of IBD were induced using indomethacin dose of 15 mg/kg orally. This study used four groups were divided into positive control group, negative control group, therapy group Solen spp. extracts dose of 100 mg/kg BW and dose of 400 mg/kg BW. Decreasing expression of TNF-α were measured using immunohistochemistry methods and analyzed by using ANOVA test followed by Honestly Significant Difference test (HSD) α=0.05. The observation of colonic histopathology were observed by microscopically and analyzed descriptively. The result showed that Solen spp. extracts repaired colonic histopathology damage on Inflammatory Bowel Disease (IBD) rats by epithelial regeneration and significantly (p<0.05) decreased expression of TNF-α. Therapeutic dose of 400 mg/kg BW decreased TNF-α expression of 55.41%. In conclusion, Solen spp. extracts have potentially as an alternative therapy in Inflammatory Bowel Disease (IBD).
