Abstract
Inflammatory Bowel Disease (IBD) merupakan penyakit inflamasi kronis pada saluran pencernaan. Salah satu penyebab IBD karena induksi indometasin, salah satu golongan obat Non-Steroidal Anti-Inflammatory Drug (NSAID) yang dapat meningkatkan produksi ROS sehingga memicu terjadinya inflamasi pada saluran pencernaan dan dapat mempengaruhi hepar. Penelitian ini menggunakan ekstrak metanol kerang pisau (Sollen spp) digunakan sebagai terapi hewan model tikus IBD. Tujuan dari penelitian ini untuk mengetahui pengaruh pemberian ekstrak metanol kerang pisau (Solen spp) terhadap aktivitas protease dan perubahan gambaran histopatologi hepar model tikus IBD. Penelitian ini menggunakan 5 kelompok tikus perlakuan, yaitu kelompok kontrol negatif, kelompok IBD, kelompok terapi 100 mg/kgBB, 400 mg/kgBB, dan 700 mg/kgBB. Penggunaan dosis indometasin adalah 15 mg/kgBB diberikan satu kali secara oral. Pemberian terapi ekstrak metanol kerang pisau dilakukan selama 14 hari. Aktivitas protease diukur dengan menggunakan spektrofotometri dan pengamatan histopatologi hepar menggunakan pewarnaan Hematoxcylin Eosin. Analisa data aktivitas protease menggunakan analisa ragam ANOVA dan uji lanjutan BNJ, serta gambaran histopatologi dianalisis secara deskriptif. Hasil penelitian menunjukkan pemberian ekstrak metanol kerang pisau dengan dosis 100, 400, dan 700 mg/kgBB secara signifikan (p<0,05) mampu menurunkan aktivitas protease, yakni berturut-turut 23,6%, 41,1%, dan 8,5%, serta memperbaiki sel hepatosit pada tikus model IBD. Kesimpulan dari penelitian ini adalah ekstrak metanol kerang pisau dapat digunakan sebagai terapi herbal pada tikus model IBD.
English Abstract
Inflammatory Bowel Disease (IBD) is an inflammatory chronic disease of the gastrointestinal tract. Indomethacin is a type of Non-Steroidal Anti-Inflammatory Drug (NSAID) that can induce IBD. Indomethacin can lead inflammation of the gastrointestinal tract and can affect the liver through ROS produced. The Solen spp methanolic extract were used as therapy agent on IBD rats. The aim of this research was to study the potential of Solen spp methanolic extract that could affect to protease activity and hepar histopathology on IBD rats. Five groups of rats which were used in this research were negative control group, IBD groups, therapy group of 100 mg/kgBW, 400 mg/kgBW, and 700 mg/kgBW. The IBD rats were induced by 15 mg/kgBW dose of indomethacin orally. Solen spp methanolic extract therapy were conducted for 14 days. Protease activity were determined using spectrofotometry and histophatological liver were evaluated with Hematoxcylin Eosin staining method. Data of protease activity were analyzed using Analysis Of Variance (ANOVA) and HSD test and histophatological of liver were analyzed descriptively. The result showed that Solen spp methanolic extract therapy decreased protease activity significantly (p<0.05). The 100 mg/kgBW, 400 mg/kgBW, and 700 mg/kgBW dose if Solen spp methanolic extract could decrease protease activity to be 23.6%, 41.1% and 8.5% respectively. It also repaired cell hepatocytes damage in IBD rats. It can be concluded that Solen spp methanolic extract can be used as herbal therapy on IBD rats.
