Abstract

Inflammatory Bowel Disease (IBD) Merupakan Suatu Kondisi Inflamasi Kronik Di Saluran Pencernaan. Inflamasi Tersebut Dapat Disebabkan Oleh Keberadaan Bakteri Patogen Maupun Penggunaan Non Steroid Anti Inflamatory Drugs (NSAID) Dalam Jangka Panjang Seperti Indometasin. Ekstrak Etanol Akar Seledri Yang Memiliki Kandungan Flavonoid Diketahui Mampu Menjadi Alternatif Terapi Kasus IBD. Penelitian Ini Bertujuan Untuk Mengetahui Kemampuan Ekstrak Etanol Akar Seledri Dalam Menurunkan Kadar Malondialdehida (MDA) Dan Memperbaiki Kerusakan Histopatologi Ileum Tikus IBD Hasil Induksi Indometasin. Penelitian Ini Menggunakan Tikus (Rattus Norvegicus) Jantan Strain Wistar Berumur 8-12 Minggu, Berat Badan 150-200 Gram Sebagai Hewan Coba. Terdapat 4 Kelompok Perlakuan, Yaitu Kelompok Kontrol, Kelompok IBD, Kelompok Terapi Ekstrak Etanol Akar Seledri Dosis 0,1 G/Kgbb Dan 0,3 G/Kgbb. Kadar MDA Diukur Dengan Uji Thiobarbituric Acid (TBA) Dan Gambaran Histopatologi Ileum Menggunakan Pewarnaan Hematoksilin-Eosin (HE). Data MDA Dianalisis Dengan ANOVA Dan Perbaikan Gambaran Histopatologi Dianalisa Secara Deskriptif. Hasil Penelitian Menunjukkan Terjadi Penurunan Kadar MDA Secara Signifikan (P<0,05) Pada Tikus Yang Diterapi Ekstrak Etanol Akar Seledri. Penurunan MDA Terbaik Sebesar 44% Pada Kelompok Terapi Dosis 0,3 G/Kgbb Dan Terjadi Regenerasi Sel Epitel Yang Ditandai Dengan Jarak Vili Yang Kembali Merapat. Kesimpulan Dari Penelitian Ini Adalah Ekstrak Etanol Akar Seledri Dapat Digunakan Sebagai Bahan Terapi Pada Tikus Model IBD

English Abstract

Inflammatory Bowel Disease (IBD) Is Gastrointestinal Tract Chronic Inflammation. It Is Caused By Pathogenic Bacterial Infection And NSAID Side Effects Consumption Such As Indomethacine. Ethanol Extract Of Celery Root Contains Flavonoid As An Antioxidants That Have Possibility Use As Alternative Therapy For IBD. The Aim Of This Research Was To Know The Effect Of Flavonoid In Ethanol Extract Of Celery Root As A Therapy For IBD Rats. The Research Observed Ethanol Extract Of Celery Root Effects On MDA Level And Ileum Histopathology On IBD Rats. The Experiment Used Male Rats (Rattus Norvegicus) Aged 8-12 Weeks And 150-200 G Weight As An IBD Model. The Research Consisted Of Control Group, IBD Group, And Two Therapy Group With Dose Of 0.1 G/Kgbw And 0.3 G/Kgbw. The MDA Level Was Determined By Thiobarbituric Acid Assay (TBA) And Ileum Histopathology Was Stained With Hematoxillin-Eosin (HE). The Data Of MDA Levels Were Analysed With ANOVA And Ileum Histopathology Was Analysed Descripitively. The Result Showed That Both Of Therapy Could Decrease MDA Level Significantly (P<0.05). The Best Result Was Obtained By Ethanol Extract Of Celery Root With Dose Therapy Of 0,3 G/BW That Could Decrease MDA Level To Be 44% And Repairing Of Ileum Histopathology That Confirmed By Epitel Regeneration And Villi Distance Remodelling. The Conclusion Of This Research Was Ethanol Extract Of Celery Root Could Be Used As Therapy For IBD.
