Abstract
Fibrosis ginjal merupakan akumulasi abnormal Extra Cellular Matrix (ECM) pada jaringan ginjal yang dapat merubah struktur ginjal normal. Penurunan ekspresi E-cadherin akan menyebabkan EMT (Epithelial to Mesenchymal Transition) sehingga terjadi peningkatan matriks ekstra seluler yang akan memicu terjadinya fibrosis ginjal. Fibrosis ginjal adalah manifestasi akhir dari Chronic Kidney Disease (CKD). Penelitian ini bertujuan untuk mengetahui potensi vitamin E sebagai terapi fibrosis ginjal terhadap ekspresi e-chaderin dan histopatologi glomerulus ginjal. Tikus model fibrosis ginjal disiapkan dengan injeksi streptokinase secara intravena pada vena coccygea dengan dosis 6000 IU/ekor tikus. Streptokinase diinduksikan sebanyak tiga kali dengan rentang waktu lima hari. Tikus dibagi menjadi lima kelompok perlakuan, yaitu kelompok kontrol positif , kelompok fibrosis ginjal, kelompok terapi vitamin E dosis 200 mg/kg BB, 300 mg/kg BB, dan 400 mg/kg BB. Ekspresi e-chaderin diamati dengan metode Imunohistokimia dan dihitung menggunakan software Axiovision serta dianalisis dengan ANOVA. Pengamatan histopatologi glomerulus ginjal dengan metode HE (Hemaktosilin Eosin) menggunakan mikroskop dan dianalisis deskriptif. Hasil penelitian menunjukkan pemberian vitamin E menaikkan ekspresi e-chaderin secara signifikan (P<0,05) dan perbaikan histopatologi glomerulus ginjal yang ditandai perbaikan capsula bowman, dan tidak adanya glumerosclerosis dan hemorraghi pada glomerulus. Dosis 400 mg/kg BB adalah dosis efektif untuk terapi fibrosis ginjal. Kesimpulan dari penelitian ini adalah pemberian vitamin E dapat digunakan sebagai terapi pada tikus model fibrosis ginjal berdasarkan ekspresi e-chaderin dan gambaran histopatologi glomerulus ginjal.
English Abstract
Renal fibrosis is characterized by the abnormal accumulation of Extra Cellular Matrix (ECM) that affected normal kidney structures. The decline in the expression of e-cadherin will cause EMT (Epithelial to Mesenchymal Transition) so that the increase in extra cellular matrix will trigger renal fibrosis. Renal fibrosis is end manifestation from Chronic Kidney Disease (CKD). Vitamin E is an antioxidant will tested as renal fibrosis therapy. The purpose of this research was to study the potential of vitamin E affect to e-chaderin expression and renal glomerulus histopathology. Renal fibrosis rats were prepared by intravenously injection of streptokinase with dose of 6000 IU, it was induced three times with five days interval. Animal models were divided into five groups were control group, renal fibrosis group, and three groups with therapy of vitamin E with dose of 200 mg/kg BW, 300 mg/kg BW, and 400 mg/kg BW. E-chaderin expressions were observed by Imunohistokimia method and determined by axiovision software and were analyzed by ANOVA. Histophatological observations of renal glomerulus were observed HE (Hemaktosilin Eosin) microscopicaly and were analyzed descriptively. The result showed that vitamin E therapy significantly (P<0.05) increased e-chaderin expressions. Moreover it also repaired renal glomerulus damage in renal fibrosis rats were marked by repairing of capsula bowman and absence of glumerosclerosis and hemorraghi on glomerulus. The dose of 400 mg/kg BW is a best therapy for renal fibrosis rats. It can be concluded that vitamin E have possibility as therapy on renal fibrosis rats based on echaderin expression and renal glomerulus histopathology.
