Abstract

Hipertiroidisme adalah suatu kondisi dimana kelenjar tiroid memproduksi hormon tiroksin (T4) dan triiodotironin (T3) secara berlebihan terutama disebabkan oleh hiperfungsi kelenjar tiroid yang menyebabkan thyrotoxicosis. Penyebab hipertiroidisme diketahui oleh beberapa faktor salah satunya faktor lingkungan yaitu ion iodida dalam jumlah berlebih atau dengan induksi Natrium Iodida (NaI). Penelitian ini bertujuan untuk mempelajari gambaran histopatologi ginjal serta ekspresi Thyroid Hormon Reseptor Alfa 1 (TR-α1) pada ginjal tikus (Rattus norvegicus) model hipertiroidisme hasil induksi NaI. Hewan coba yang dipakai yaitu tikus (Rattus norvegicus) betina usia 8-12 minggu yang dibagi dalam 5 kelompok yaitu kelompok kontrol (A) dan kelompok hipertiroidisme yang diinduksi NaI 0,05% selama 2 minggu (B), 4 minggu (C), 6 minggu (D), 8 minggu (E). Ekspresi TR-α1 diamati menggunakan metode imunohistokimia yang dianalisis secara statistik kuantitatif menggunakan uji One-way Annova dan dilanjut dengan uji BNJ (tuckey) α = 0,05%. Gambaran histopatologi ginjal dengan pewarnaan Hematoxilin-Eosin diamati secara deskriptif kualitatif. Hasil penelitian ini menunjukkan ekspresi TR-α1 berbeda nyata antar perlakuan (p<0,05). Ekspresi TR-α1 pada masing-masing kelompok perlakuan hipertiroidisme mengalami peningkatan dibanding dengan kelompok kontrol. Peningkatan TR-α1 paling signifikan ditunjukan oleh kelompok (E) sebesar 477,37%. Gambaran histopatologi ginjal pada kelompok hipertiroidisme menunjukan adanya hipertrofi sel tubulus, hipertrofi sel mesengial dan podocytes, glomerulosclerosis dan terjadi piknosis sel tubulus distal. Dari hasil penelitian ini dapat disimpulkan bahwa hipertiroidisme hasil induksi NaI dengan waktu inkubasi yang semakin lama akan meningkatkan ekspresi TR-α1 dan menyebabkan perubahan histopatologi ginjal tikus model hipertiroidisme.

English Abstract

Hyperthyroidism is a condition when the thyroid gland over production of thyroxine (T4) and triiodothyronine (T3) hormones, especially is caused by tiroid glands hiperfunction that causes thyrotoxicosis. Hyperthyroidism caused by several factors which one is environmental iodide ions in large amount or with induced Sodium Iodide (NaI). This study was aimed to learn renal histopathology of rat (Rattus norvegicus) hyperthyroidism models and Thyroid Hormone Receptor expression Alpha 1 (TR-α1) in the rat model of renal hyperthyroidism. The experiment was used male (Rattus norvegicus) age of 8-12 weeks were divided into 5 groups: control group (A), and hyperthyroidism group-induced NaI 0.05% for 2 weeks (B), 4 weeks (C), 6 weeks (D), 8 weeks (E). The expression TR-α1 observed by using immunohistochemistry and analyzed statistically using a quantitative with one-way Annova test then Tuckey. Renal histopathology with Hematoxilin-Eosin staining observed in descriptive qualitative. The results of this study showed significant difference of TR-α1 expression between treatments (p <0.05). Hyperthyroidism group showed the increasing of TR-α1 expression compared with control group. Improvement of the most significant TR-α1 showed by (E) group as much as 477.37% with an average TR-α1 expression 0.791 ± 0.02. Renal histopathology of hyperthyroidsm group showed a tubular hypertrophy, mesengial cell and podocytes hypertrophy, glomerulosclerosis and piknosis in tubules. From the result can be conclude that hyperthyroidism induction of NaI with the longer incubation time can increased the expression of TR-α1 and renal histopathological structure changes in rat models of hyperthyroidism.
