Abstract
Salmonellosis merupakan penyakit zoonosis yang disebabkan oleh infeksi Salmonella sp. yang mampu menginfeksi manusia maupun hewan melalui makanan dan minuman yang tercemar. Ekstrak etanol kulit biji jambu mete (Anacardium occidentale) mengandung zat aktif asam anakardat, kardol dan kardanol berpotensi sebagai antimikroba dan antioksidan. Tujuan penelitian ini adalah untuk mengetahui potensi terapi ekstrak etanol kulit biji jambu mete (Anacardium occidentale) terhadap ekspresi iNOS dan histopatologi kolon pada tikus (Rattus norvegicus) yang diinfeksi Salmonella sp. dosis 108 CFU/ml/ekor secara per oral. Penelitian ini menggunakan 4 kelompok yang terbagi atas kelompok kontrol negatif, kontrol positif, kelompok terapi ekstrak kulit biji jambu mete dosis 50 mg/kg BB dan 100 mg/kg BB. Penurunan ekspresi iNOS diamati dengan metode imunohistokimia yang dianalisa menggunakan software axiovision dan dilakukan analisa ANOVA serta dilanjutkan uji BNJ (Beda Nyata Jujur) α= 0,05. Gambaran histopatologi kolon diamati dengan mikroskop cahaya dan dilakukan analisis secara deskriptif. Hasil penelitian menunjukkan bahwa pemberian ekstrak kulit biji jambu mete menurunkan ekspresi iNOS secara signifikan (p<0,05) antar kelompok perlakuan, kelompok dengan dosis terapi 100mg/kg BB adalah dosis terbaik menurunkan ekspresi iNOS sebesar 65,03% Pemberian terapi dosis 100 mg/kg BB memperbaiki histopatologi kolon tikus yang diinfeksi Salmonella sp. Penelitian ini menyimpulkan bahwa terapi ekstrak etanol kulit biji jambu mete dapat menurunkan ekspresi iNOS dan memperbaiki histopatologi kolon pada tikus (Rattus norvegicus) yang diinfeksi Salmonella sp.
English Abstract
Salmonellosis is a zoonotic disease caused by Salmonella sp. which are able to infect human and animals through contaminated of food and drink. The cashew nut (Anacardium occidentale) shell ethanol extract contain an active substance that has potentially as antimicrobial and antioxidant such as anacardic acid, cardol and cardanol. The purpose of this research was to determine the therapeutic effect of cashew nut (Anacardium occidentale) shell ethanol extract against the Inducible Nitrit Oxide Synthase (iNOS) expression and colon histopathology in rat (Rattus norvegicus) infected by 108 dose of Salmonella sp. CFU/mL orally. This study divided into 4 groups : negatif control group, positive control group, and induced by dose of 50 mg/BW and 100 mg/BW of ethanol extract of cashew nut shell (pericarp). Decreasing of iNOS expression was observed by immunohistochemistry method and measured use AxioVision software then analyzed with ANOVA test followed by (Honestly Significant Difference)HSD test (α = 0.05). Colon histopathology were observed by microscope and analyzed descriptively. The results showed that cashew nut shell extract decrease the inducible nitrit oxide synthase (iNOS) expression significantly (p <0.05), group with therapy dose of 100mg/BW was the best dose to decrease iNOS expression showed by improvement of 65.03% and repaired colon histopathology damage in rats infected by Salmonella sp. The conclusion of this study is that therapy of cashew nut (Anacardium occidentale) shell ethanol extract is able to decrease expression of iNOS and repair colon histopatology on rat (Rattus norvegicus) infected by Salmonella sp.
