Abstract

Diabetes Melitus (DM) merupakan suatu penyakit metabolit dengan karakteristik hiperglikemia yang terjadi karena kelainan sekresi insulin, kerja insulin, atau kedua-duanya. Daun tanaman nangka (Artocarpus heterophyllus) direkomendasikan sebagai obat antidiabetes yang mampu menurunkan kadar glukosa darah. Penelitian ini dilakukan dengan tujuan untuk mengetahui potensi ekstrak etanol daun nangka (Artocarpus heterophyllus) dalam pengobatan DM melalui penurunan kadar malondialdehida (MDA) dan aktivitas enzim superoksida dismutase (SOD) serum. Tikus model DM disiapkan dengan induksi streptozotocin (STZ) dengan dosis multiple low dose-streptozotocin (MLD-STZ) 20mg/KgBB selama 5 hari berturut-turut secara intraperitoneum. Penelitian ini menggunakan lima kelompok perlakuan yaitu kelompok kontrol negatif, kontrol DM, kelompok terapi daun nangka dosis 50 mg/kgBB, kelompok terapi daun nangka dosis 100 mg/kgBB, dan kelompok terapi daun nangka dosis 200 mg/kgBB. Kadar MDA diukur menggunakan metode Thiobarbituric Acid (TBA) dan akitivitas enzim Superoksida Dismutase (SOD) diukur menggunakan kit SOD (Cat. K335-100, Biovision). Hasil penelitian menunjukkan pemberian terapi ekstrak etanol daun nangka (Artocarpus heterophyllus) secara signifikan (p<0,05) mampu menurunkan kadar MDA dan mampu meningkatkan aktivitas enzim superoksida dismutase (SOD). Dosis efektif pemberian ekstrak etanol daun nangka (Artocarpus heterophyllus) adalah sebesar 200mg/KgBB.

English Abstract

Diabetes mellitus is a metabolic disease with hyperglycemia because of insulin abnormality, insulin function, or both. Jackfruit leaves (Artocarpus heterophyllus) recommended as antidiabetic medicine because it provide hypoglicemia effect. The aim of this study was to determine the potential of the ethanol extract of jackfruit’s leaf (Artocarpus heterophyllus) in the treatment of diabetes by decrease levels of malondialdehida (MDA) and activity of superoxide dismutase (SOD) of serum. Rat DM model prepared with induction streptozotocin (STZ) with multiple low dose-streptozotocin (MLD-STZ) 20mg/KgBW for consecutive 5 days in intra peritoneum. This study used five treatment groups those were negative control group, DM control, group of jackfruit leaf’s therapy dose 50 mg/kg, group of jackfruit leaves therapy dose 100 mg/kg, and group of jackfruit leafs therapy dose 200 mg/kg. MDA levels were measured using by the thiobarbituric acid (TBA) and superoxide dismutase (SOD) activity was measured with SOD kit (Cat. K335-100, Biovision). The results showed that therapy of jackfruit leaves ethanol extract (Artocarpus heterophyllus) could decrease MDA levels and increase superoxide dismutation (SOD) activity significantly (p<0,05). Effective dose of ethanol extract of jackfruit’s leaf (Artocarpus heterophyllus) was 200mg/KgBB.
