Abstract

Hiperkolesterolemia disebabkan oleh peningkatan kadar LDL melebihi batas normal. Tingginya LDL dalam darah mudah masuk ke dalam intima pembuluh darah dan menstimuli terbentuknya radikal bebas. Beberapa penelitian telah membuktikan bahwa ekstrak tanaman Alfalfa memiliki kandungan bioaktif berupa flavonoid yang dapat mencegah peningkatan kadar kolesterol dalam darah. Tujuan penelitian ini adalah untuk mengetahui pengaruh terapi ekstrak tanaman Alfalfa (Medicago sativa) terhadap kadar Malondialdehyde (MDA) dan gambaran histopatologi aorta tikus hiperkolesterolemia. Penelitian ini menggunakan tikus (Rattus norvegicus) jantan strain Wistar umur 8-12 minggu dengan berat badan 160-200 g yang dibagi dalam lima kelompok perlakuan yaitu kelompok kontrol negatif, hiperkolesterolemia, dan kelompok terapi ekstrak tanaman Alfalfa masing-masing0,36 mg/200g BB, 0,72 mg/200g BB dan 1,08 mg/200g BB. Pembuatan hewan model hiperkolesterolemia dengan induksi diet hiperkolesterol dan terapi diberikan selama 14 hari. Pengukuran kadar Malondialdehide (MDA) menggunakan teknik Thiobarbituric Acid (TBA) dan pembuatan preparat histopatologi aorta dengan pewarnaan Hematoksilin Eosin (HE). Data yang diperoleh dianalisis menggunakan SPSS versi 16.0 dengan analisis ragam One Way Analisis Of Variance (ANOVA) dan dilakukan analisis lebih lanjut dengan uji Tukey5%. Hasil penelitian menunjukkan MDA dan jumlah foam cell histopatologi aorta berbeda nyata (p<0,05) antar perlakuan. Dosis terapi 1,08 mg/200g BB merupakan dosis optimum dalam menurunkan kadar MDA (62,2%) dan jumlah foam cell (82%) aorta hewan model tikus hiperkolesterolemia. Kesimpulan dari penelitian ini adalah ekstrak tanaman alfalfa dapat dijadikan salah satu alternatif pengobatan hiperkolesterolemia.

English Abstract

Hypercholesterolemia caused by elevated levels of LDL blood. High LDL blood levels easily into the intima of blood vessels and stimulate the formation of free radicals. The several studies have shown that extractof Alfalfa plants contain flavonoid bioactive which can prevent the increasing of cholesterol levels in the blood.The aim of this research was to study therapeutic effect of chlorophyll from Alfalfa (Medicago sativa) on the malondialdehyde (MDA) levels and aorta’s histopathology of hypercholesterolemic rats. This research used male rats, aged 8-12 weeks, weight of 160-200 g, which were divided into five groups, were control negative, hypercholesterolemia, extract of Alfalfa therapy dose of 0.36 mg/200g BW, 0.72 mg/200g BW and 1.08 mg/200g BW were given for 14 days. The hypercholesterolemic rats were prepared by administered hypercholesterolemia diet and chlorophyll of Alfalfa therapy for 14 days. The MDA levels measured using the Thiobarbituric Acid (TBA) methode and aortic histopathologywas analyzed microscopically with hematoxylin eosin staining (HE). The data were analyzed by SPSS 16.0 version using one way Analysis of Variance (ANOVA), continued by Tukey test 5%. The result showed that the MDA level and foam cell levels from each group was significantly different (p<0.05). The extract of Alfalfa therapy dose of 1.08mg/200gBW was the optimum dose for decreasing MDA level (62.2%) and foam cell levels (82%) in aorta of hypercholesterolemic rats. It could be concluded, the extract of Alfalfa therapy could be used as alternative therapy for hypercholesterolemia.
