Abstract

Hiperkolesterolemia merupakan kondisi adanya peningkatan kolesterol dalam darah. Keadaan hiperkolesterolemia memicu peningkatan radikal bebas dan stres oksidatif. Yoghurt susu kambing diketahui memiliki kandungan peptida α laktobumin dan α kasein yang berpotensi sebagai antioksidan. Tujuan penelitian ini adalah untuk mengetahui pengaruh yoghurt susu kambing untuk menurunkan kadar MDA dan ekspresi TNFα ginjal tikus (Rattus norvegicus) model hiperkolesterolemia. Hewan coba yang digunakan dalam penelitian ini tikus (Rattus norvegicus) jantan umur 10-12 minggu, dengan rata-rata bobot badan 100- 150 g, dibagi menjadi 5 kelompok perlakuan: kelompok kontrol, kelompok hiperkolesterolemia, dan 3 kelompok terapi dengan dosis 300mg/kg BB, 600 mg/kg BB, dan 900 mg/kg BB. Pemberian diet tinggi kolesterol berupa pakan yang mengandung minyak babi, asam kholat dan kuning telur puyuh rebus diberikan melalui metode sonde lambung dan diberikan selama 2 minggu. Terapi yoghurt susu kambing dalam bentuk freeze dry dengan konsentrasi 109 cfu/mL diberikan selama 4 minggu. Kadar MDA diukur dengan metode TBA dan ekspresi TNFα diamati dengan metode imunohistokimia. Hasil penelitian menunjukan bahwa terapi yoghurt susu kambing secara signifikan (p<0,05) mampu menurunkan kadar MDA dan ekspresi TNFα. Dosis 900 mg/kg BB merupakan dosis efektif dengan persentasi penurunan kadar MDA sebesar 42,02% dan ekpresi TNFα sebesar 71,65 %. Kesimpulan dari penelitian ini terapi yoghurt susu kambing dapat digunakan sebagai alternatif terapi hiperkolesterolemia.

English Abstract

Hypercholesterolemia is a condition of high cholesterol level in blood. Hypercholesterolemia triggers increasing of free radical and oxidative stress. Goat milk yogurt contains bioactive peptide such as α lactobumin and α casein that used as an antioxidant. The aim of this research was to determine the potency of goat milk yogurt as hypercholesterolemia therapy based on MDA levels and TNFα expression in kidney of hypercholesterolemic rats. This research used male rats (Rattus norvegicus) with range of age 10-12 weeks and weight average of 100-150g which divided into 5 groups: control group, hypercholesterolemia group, and goat milk yoghurt therapy of 300 mg/kg BW, 600 mg/kg BW, and 900 mg/kg BW. Hypercholesterolemic condition induced by high fat diet contains of lard, cholid acid, and boiled quail egg yolks during 2 weeks orally. The therapy used goat milk yogurt concentration of 109 cfu/mL during 4 weeks at dosage of 300 mg/kg BW, 600 mg/kg BW, and 900 mg/kg BW. The MDA levels were measured by TBA method and TNF expression observed by immunohistochemistry. The result showed that goat milk yogurt therapy significantly (p<0.05) decreased MDA levels and TNF expression. The dose of 900 mg/kg BW was an effective dose that could decreased MDA levels to 42.02% and expression of TNF to 71.65% compared to control group. These result provide that goat milk yogurt could be used as an alternative therapy for hypercholesterolemia.
