Abstract

Hiperkolesterol merupakan penyakit gangguan metabolisme kolesterol yang disebabkan oleh kadar kolesterol dalam darah melebihi batas normal. Salah satu alternatif yang dapat menurunkan kadar kolesterol dalam darah ialah ekstrak tanaman Alfalfa (Medicago sativa). Tujuan penelitian ini untuk mengetahui pengaruh pemberian klorofil dari tanaman Alfalfa (Medicago sativa) terhadap kadar kolesterol total dan aktifitas enzim lipoprotein lipase (LPL) pada tikus yang model hipekolesterol. Penelitian experimental ini menggunakan rancangan acak lengkap dengan pretest-posttest control group design. Hewan uji dibagi dalam 5 kelompok perlakuan. Kelompok P1 kontrol negatif diberi pakan standart. Kelompok P2 kontrol positif diberi pakan hiperkolesterol tanpa terapi ektrak tanaman alfalfa. Kelompok P3, P4, dan P5 diberi pakan hiperkolesterol dan mendapat terapi ekstrak tanaman alfalfa dengan dosis 0,36mg/ekor, 0,72 mg/ekor, dan 1,08 mg/ekor selama 14 hari. Analisis yang digunakan dalam penelitian ini adalah One Way Analisis Of Variance (ANOVA) dengan uji lanjutan Tukey. Hasil penelitian menunjukkan bahwa terapi ekstrak tanaman alfalfa dapat menurunkan kadar kolesterol total dan meningkatkan aktivitas enzim LPL secara signifikan pada tikus (Rattus norvegicus) model hiperkolesterolemia. Hasil penelitian menunjukkan terapi ekstrak tanaman alfalfa (Medicago sativa) dengan dosis 1,08 mg/ekor merupakan dosis optimal dalam menurunkan kadar kolesterol, dengan penurunan kadar kolesterol sebesar 57,51% dan dosis 0,72 mg/ekor merupakan dosis optimal dengan meningkatkan aktifitas enzim LPL dalam kenaikan aktivitas enzim LPL sebesar 201,07%. Dapat disimpulkan bahwa ekstrak tanaman alfalfa dapat digunakan sebagai terapi hiperkolesterol yang ditandai dengan peningkatan aktivitas enzim LPL dan menurunkan kadar kolesterol total.

English Abstract

Hypercholesterol is a cholesterol metabolism disorder disease caused by high cholesterol level in the blood. One alternative to reduce blood-based cholesterol level is treatment with the extract of Alfalfa Plant (Medicago sativa). The objective of this research is to the investigase influence of treatment with extracted Alfalfa Plant (Medicago sativa) on total cholesterol level and activity of lipase lipoprotein enzyme (LPL) in hypercholesterol model rat. This experimental design were completely randomized pretest-posttest control group design. Rats is assigned into 5 treatment groups with complete random planning method. Group 1, negatif control was given normal diet. Group 2, positif control was received hypercholesterol diet without alfalfa therapi. Group 3,4,and 5 were treated with hypercholesterol diet with 0.36 mg/rat, 0.72 mg/rat, and 1.08 mg/rat of alfalfa therapi respecsively for 14 day. Total cholesterol level was measured by using CHOD-PAP and activity of LPL was measured by using alkalimetic titration. The data were analyzed by usisng One Way Analysis of Variance (ANOVA) followed by advance test with Post Hoc Test. The Result showed that therapy with extract of Alfalfa Plant (Medicago sativa) at dose 1.08 mg/rat was an optimal dose that lowered 57,51% cholesterol level and dose 0.72 mg/rat was an optimal dose that increasing the activity of LPL enzyme levels with 201,07% increase activity of LPL. It was concluded that the extract of Alfalfa Plant (Medicago sativa) can be used for the treatment of hypercholesterolemia which indicated by the increased of LPL activity and reduce of total cholesterol
