Abstract
Rheumatoid arthritis (RA) adalah penyakit autoimun yang ditandai dengan inflamasi kronik pada persendian. Tujuan penelitian ini adalah untuk mengetahui potensi pemberian ekstrak etanol daun anting-anting (Acalypha indica L.) terhadap ekspresi inducible nitric oxide synthase (iNOS) dan perbaikan gambaran histopatologi jaringan sendi metatarsophalangeal pada tikus hasil induksi complete freud’s adjuvant (CFA). Penelitian ini terdiri dari empat kelompok perlakuan yaitu kontrol negatif, kontrol positif, terapi ekstrak etanol daun anting-anting dosis 400 mg/kg BB dan terapi ekstrak etanol daun anting-anting dosis 1000 mg/kg BB. Parameter yang diamati dalam penelitian ini adalah ekspresi iNOS yang diamati dengan metode imunohistokimia kemudian dilakukan analisis ragam secara kuantitatif dengan ANOVA dan dilakukan analisis lebih lanjut dengan uji Tukey (α = 0,05). Gambaran histopatologi jaringan sendi metatarsophalangeal dilakukan dengan pewarnaan Hematoxylin Eosin (HE) kemudian diamati dengan mikroskop cahaya perbesaran 400X dan dianalisa secara kualitatif. Hasil penelitian menunjukkan terapi ekstrak etanol daun anting-anting dapat menurunkan ekspresi iNOS secara signifikan (p<0,05) antar kelompok perlakuan. Penurunan ekspresi terbaik pada pemberian terapi dosis 1000 mg/kg BB sebesar 35,3%. Histopatologi jaringan sendi metatarsophalangeal menunjukkan adanya perbaikan pada kartilago dan susunan kondrosit.
English Abstract
Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic inflammation of the joints. The aim of this study was determine the potency of ethanol extract of the Acalypha indica leaves against inducible nitric oxide synthase (iNOS) expression and the improvement of metatarsophalangeal joint tissue histopathological in rats which were induced by complete freuds adjuvant (CFA). This study consisted of four treatment groups were negative control, positive control, therapy ethanol extract of Acalypha indica leaves which dose of 400 mg/kg BW and therapy ethanol extract of the leaves of Acalypha indica dose of 1000 mg/kg BW. The parameters observed in this study were the expression of iNOS observed by immunohistochemical methods and then performed a quantitative analysis of variance with ANOVA and carried out further analysis with Tukey test (α = 0.05). Metatarsophalangeal joint tissue histopathology was stained with Hematoxylin eosin (HE), and then it was observed with a light microscope magnification of 400X then it was analyzed qualitatively. The results of this study showed that ethanolic extract of Acalypha indica leaves with therapeutic dose of 400 mg/kg BW and 1000 mg/kg BW could descrease the expression of iNOS significantly (p <0.05) between the treatment groups. The ethanolic extract of Acalypha indica leaves therapeutic dose of 1000 mg/kg BW could descrease the expression of iNOS was 35.3% and metatatarsophalangeal joint tissue histopathology showed the improvement in the composition of cartilage and condrocytes of joint tissue.
