Abstract

Fibrosis Ginjal Merupakan Hilangnya Fungsi Ginjal Karena Terdapat Akumulasi Fibroblas Dan Matriks Protein Pada Jaringan Ginjal. Aktifitas Radikal Bebas Dapat Diturunkan Dengan Antioksidan. Salah Satu Antioksidan Adalah Vitamin E Yang Di Yakini Dapat Digunakan Sebagai Terapi Fibrosis Ginjal. Penelitian Ini Bertujuan Untuk Mengetahui Pengaruh Pemberian Vitamin E Terhadap Penurunan Kadar Malondialdehid (MDA) Dan Perbaikan Fibrosis Ginjal Tikus. Penelitian Ini Menggunakan Tikus (Rattus Norvegicus) Dengan Berat 120-170 Gram. Tikus Model Fibrosis Ginjal Disiapkan Dengan Injeksi Streptokinase Secara Intravena Pada Vena Coccygea Dengan Dosis 6000IU/Ekor Tikus, Sebanyak Tiga Kali Dengan Rentang Waktu Lima Hari. Tikus (Rattus Norvegicus) Dibagi Menjadi 5 Kelompok Yaitu Kelompok Kontrol, Kelompok Fibrosis Ginjal, Kelompok Fibrosis Ginjal Yang Telah Diterapi Vitamin E Dosis 200 Mg/Kg BB, 300 Mg/Kg BB, Dan 400 Mg/Kg BB. Kadar MDA Diukur Menggunakan Metode Thiobarbaturic Acid (TBA) Dianalisis Dengan ANOVA Yang Dilanjutkan Dengan Uji Tukey (Beda Nyata Jujur) Dengan (Α =5%). Pengamatan Histopatologi Tubulus Ginjal Menggunakan Mikroskop Olympus BX51 Dan Dianalisis Deskriptif. Hasil Penelitian Menunjukkan Bahwa Pemberian Vitamin E Α-Tokoferol Secara Signifikan (P<0,05) Mampu Menurunkan Kadar MDA Dan Memperbaiki Sel Epitel Pada Histopatologi Tubulus Ginjal. Dosis Vitamin E 300 Mg/Kg BB Adalah Dosis Efektif Untuk Terapi Fibrosis Ginjal. Kesimpulan Dari Penelitian Ini Adalah Pemberian Vitamin E Dapat Digunakan Sebagai Terapi Fibrosis Ginjal Berdasarkan Kadar MDA Dan Gambaran Histopatologi Tubuluslus Ginjal.

English Abstract

Renal Fibrosis Is Loss Of Kidney Function Because There Is Accumulation Of Fibroblast And Matrix Protein In Kidney Tissue. Free Radical Activity Can Be Decreased With Antioxidants. One Of Antioxidants Is Vitamin E Used As A Renal Fibrosis Therapy. The Purpose Of This Research Was To Study The Potential Of Vitamin E Toward MDA Level And Repaired Histopathology Renal Damage Of Rats (Rattus Norvegicus). Renal Fibrosis Rats Were Prepared By Intravenously Injection Of Streptokinase With Dose Of 6000 IU, For Three Times With Five Day Interval. Rats (Rattus Norvegicus) Were Divided Into 5 Groups: Control Group, Renal Fibrosis Group, And Three Groups With Therapy Of Vitamin E Dose Of 200 Mg/Kg BW, 300 Mg/Kg BW, And 400 Mg/Kg BW. The Levels Of MDA Were Determined Using Thiobarbaturic Acid (TBA) Method And Were Analyzed With ANOVA. That Continued With Tukey Test By (Α = 5%). The Histopathological Of Renal Tubules Were Observed Microscopically Olympus BX51 And Were Analyzed Descriptively. The Result Of This Research Showed That Vitamin E Therapy Significantly (P <0.05) Can Decrease The MDA Level And Repair Epithel Cells On Renal Histopathology Tubules. The Dose Therapy Vitamin E Of 300 Mg/Kg BW Was The Effective Dose On Renal Fibrosis Rats. It Can Be Concluded That Vitamin E Therapy Have Possibility As Therapy Of Renal Fibrosis Based On MDA Level And Repair Renal Tubululer Histophatology. Keywords:
