Abstract

Hipertensi merupakan peningkatan tekanan darah lebih dari 140/90 mmHg dan merupakan faktor resiko penyakit serebrovaskuler seperti stroke. Pengobatan hipertensi menggunakan obat kimiawi meningkatkan kemungkinan terjadinya efek samping. Yoghurt susu kambing dapat menjadi alternatif terapi hipertensi karena mengandung bioaktif peptida yang berfungsi sebagai antioksidan dan antihipertensi. Penelitian ini bertujuan untuk mengetahui pengaruh terapi yoghurt susu kambing terhadap kadar malondialdehida (MDA) dan gambaran histopatologi otak tikus hipertensi induksi DOCA-salt. Penelitian ini menggunakan tikus (Rattus norvegicus) strain Wistar jantan berumur 10 – 12 minggu yang dibagi dalam 5 kelompok perlakuan: normotensif, hipertensif, terapi yoghurt 300 mg/KgBB, terapi yoghurt 600 mg/KgBB dan terapi captopril 5 mg/KgBB. Induksi hipertensi dilakukan dengan injeksi deoxycorticosterone acetate (DOCA) secara subkutan disertai diet larutan NaCl 2% ad libitum. Kadar MDA otak diukur menggunakan metode Thiobarburic Acid (TBA) dan pengamatan histopatologi otak menggunakan metode pewarnaan Hematoxylin-Eosin (HE). Hasil penelitian menunjukkan bahwa kelompok terapi yoghurt dosis 300 mg/kgBB (0,281 ± 0,046 μg/ml) dan 600 mg/kgBB (D) (0,213 ± 0,077 μg/ml) menunjukkan nilai kadar MDA yang tidak berbeda nyata (P<0,05) terhadap kelompok normotensif (0,174 ± 0,062 μg/ml) namun menunjukkan nilai MDA yang berbeda nyata terhadap kelompok hipertensif (0,459 ± 0,025 μg/ml). Pemberian terapi yoghurt menunjukkan adanya perbaikan gambaran histopatologi otak tikus yang ditunjukkan dengan perbaikan endotel dan tidak ditemukannya gambaran hemoragi. Hasil penelitian menyimpulkan bahwa terapi yoghurt susu kambing dapat menurunkan kadar MDA dan memperbaiki histopatologi otak tikus model hipertensi hasil induksi DOCA-salt.

English Abstract

Hypertension is an elevation in blood pressure over 140/90 mmHg and a major cause of mortality due to its association with cerebrovascular disease such as stroke. Treatment of hypertension using chemical drugs increases the likelihood of side effects; therefore goat milk yoghurt can be used as alternative treatment due to its bioactive peptides which function as both antioxidants and antihypertensive. The present study was designed to determine dose dependent effects of goat milk yoghurt treatment on Malondialdehyde (MDA) level and histopathology of brain in Deoxycorticosterone Acetate (DOCA)-salt hypertensive rats. This study used male Wistar strain rat (Rattus norvegicus) aged 10-12 weeks which were divided into five treatment groups: normotensive, hypertensive, yoghurt therapy of 300 mg/KgBW, yoghurt therapy of 600 mg/kgBW and captopril therapy of 5 mg/KgBW. Hypertension was induced by subcutaneous injection of DOCA with ad libitum diet of NaCl 2%. Brain MDA levels were measured using Thiobarburic Acid (TBA) method and histopathology were observed using Hematoxylin-Eosin (HE) staining. The results showed that MDA levels of therapy group 300 mg/kgBW (0,281 ± 0,046 μg/ml) and 600 mg/kgBW (0,213 ± 0,077 μg/ml) was not significantly different (P < 0.05) to normotensive group (0,174 ± 0,062 μg/ml); however, it was significantly different to hypertensive group (0,459 ± 0,025 μg/ml). Yoghurt treatment demonstrated a repair in endothelial as well as reduction of hemorrhage. The current study suggests that goat milk yoghurt treatment was capable of reducing MDA levels and repair histopathological alterations in brain of DOCA-salt hypertensive rats.
