Abstract
Hipertensi merupakan keadaan peningkatan tekanan darah pada pembuluh darah yang berada diatas normal yaitu ≥ 135/90 mmHg. Hipertensi berpotensi menyebabkan kerusakan pada pembuluh darah. Hipertensi dapat terjadi akibat adanya induksi deoxycorticosterone acetate (DOCA)-salt. DOCA-salt menyebabkan peningkatan stres oksidatif sehingga berdampak pada perubahan jaringan organ aorta. Water Soluble Extract (WSE) yoghurt susu kambing mengandung bioaktif peptida yang berguna sebagai antioksidan dan antihipertensi. Penelitian ini bertujuan untuk mengetahui pengaruh terapi WSE yoghurt susu kambing terhadap kadar malondialdehida (MDA) dan gambaran histopatologi organ aorta tikus hipertensi induksi DOCA-salt. Penelitian ini menggunakan tikus jantan yang terbagi dalam 5 kelompok perlakuan. Induksi hipertensi dilakukan dengan injeksi deoxycorticosterone acetate (DOCA) secara subcutan disertai diet larutan NaCl 2% ad libitum. Hasil penelitian menunjukkan bahwa terapi WSE yoghurt susu kambing dosis 600 mg/kgBB merupakan dosis terbaik dalam menurunkan kadar MDA terhadap kelompok hipertensi (p<0,05). Gambaran histopatologi organ aorta menunjukkan ditemukannya gambaran sel otot polos normal dan tidak ditemukannya gambaran proliferasi serta sel nekrosis pada tunika media dinding organ aorta tikus terapi WSE yoghurt susu kambing. Hasil penelitian menyimpulkan bahwa water soluble extract (WSE) yoghurt susu kambing dapat menurunkan kadar malondialdehida dan dapat memperbaiki kerusakan jaringan pada organ aorta dengan memperbaiki proliferasi dan nekrosis sel otot polos pada tunika media dinding organ aorta.
English Abstract
Hypertension is the increasing of blood pressure in blood vessels that are above ≥ 135/90 mmHg. Hypertension is a potential cause of blood vessels damage. Hypertension can occur due to the induction of deoxycorticosterone acetate (DOCA)-salt. DOCA-salt increased oxidative stress thus it can cause vascular tissue change in aorta. Water Soluble Extract (WSE) goat milk yogurt contains bioactive peptides that have usefulness as antioxidant and antihypertensive. This research aim was to know about therapeutic effects of water soluble extract (WSE) goat milk yogurt to malondialdehyde (MDA) levels and histopathology of aorta rat (Rattus norvegicus) in hypertension induce DOCA-salt. This research used male rats which were divided into 5 groups of treatment. Hypertension induced by deoxycorticosterone acetate (DOCA) subcutaneous injection and 2% NaCl diet ad libitum. The results showed that WSE goat milk yogurt 600 mg/kgBB was the best dose to reduce MDA levels of hypertension group (p<0.05). Histopathology of aorta WSE goat milk yogurt therapy showed a normal smooth muscle cells and there were no proliferation and necrosis cell of the aortic tunica media. The research concluded that WSE goat milk yogurt could lower the malondialdehida levels and repair tissue damage in aortic tunica media by repairing proliferation and necrosis cell
