Abstract
Hipertensi dicirikan dengan peningkatan tekanan darah sistolik > 140 mmHg dan diastolik > 90 mmHg, dapat menyebabkan hipertrofi ventrikel kiri, stroke dan gagal ginjal. Hipertensi diinduksi dengan deoxycorticosterone acetate (DOCA)-salt, menyebabkan retensi natrium dan air yang berujung terjadinya stres oksidatif yang berdampak pada kerusakan jantung. Water soluble extract (WSE) yogurt susu kambing diketahui mengandung peptida bioaktif dengan aktivitas antihipertensi dan antioksidan. Tujuan penelitian ini adalah mengetahui pengaruh terapi WSE yogurt susu kambing terhadap kadar MDA dan gambaran histopatologi jantung. Parameter yang diukur adalah kadar MDA dengan metode thiobarbituric acid (TBA) dan gambaran histopatologi jantung dengan pewarnaan hematoxylin eosin. Penelitian ini menggunakan 20 ekor tikus wistar jantan yang dibagi dalam 5 kelompok perlakuan yaitu kelompok kontrol, kelompok hipertensi, kelompok terapi captopril, kelompok WSE yogurt susu kambing 300 mg/kg BB dan 600 mg/kg BB. Hasil penelitian menunjukkan kadar MDA kelompok hipertensi (0,651±0,049 μg/mL) berbeda nyata (p<0,05) dibandingkan kontrol (0,306±0,069 μg/mL). Kadar MDA terapi WSE yogurt susu kambing 300 mg/kg BB (0,436±0,065 μg/mL) tidak berbeda nyata dibandingkan terapi captopril (0,524±0,047 μg/mL) dan terapi WSE yogurt susu kambing dosis 600 mg/kg BB (0,377±0,045 μg/mL) tidak berbeda nyata dibandingkan kontrol. Gambaran histopatologi jantung menunjukkan perbaikan myosit dan struktur myofibril. Dari hasil penelitian dapat disimpulkan bahwa dosis terapi WSE yogurt susu kambing 600 mg/kg BB menunjukkan dosis yang paling efektif menurunkan kadar MDA dan perbaikan gambaran histopatologi jantung.
English Abstract
Hypertension is characterized with a systolic blood pressure over 140 mmHg and diastolic blood pressure over 90 mmHg. It leads to left ventricle hypertrophy, stroke and renal failure. Hypertension induced by deoxycorticosterone acetate (DOCA)-salt caused sodium and water retention which lead to oxidative stress, that impact the heart damage. Water soluble extract (WSE) of goat milk yogurt containing bioactive peptides which have antihypertensive and antioxidant activities. The aim of this study was to study effect WSE of goat milk yogurt therapy to malondialdehyde (MDA) levels and histopathology of the heart. Parameters measured were the MDA levels with thiobarbituric acid method (TBA) and histopathology of the heart with hematoxylin eosin staining. This study used 20 male wistar rats that were divided into 5 groups, namely control group, hypertension group, captopril therapy group, WSE of goat milk yogurt 300 mg/kg BW and 600 mg/kg BW therapy group. The results showed MDA levels of hypertension group (0.651±0.049 μg/mL) was significantly different (p<0.05) compare to control group (0.306±0.069 μg/mL). MDA levels of WSE goat milk yogurt 300 mg/kg BW therapy group (0.436±0.065 μg/mL) was not significantly different compare to captopril therapy group (0.524±0.047 μg/mL) and WSE of goat milk yogurt 600 mg/kg BW therapy group (0.377±0.045 μg/mL) was not significantly different compare to control group. Histopathology observation of the heart showed repairing of myocyte and myofibrile structure. Therapeutic dose of 600 mg/kg BW of WSE goat milk yogurt was the most effective dose to decrease levels of MDA and repair heart histopathology.
