Abstract
Inflammatory Bowel Disease (IBD) Merupakan Penyakit Pada Saluran Pencernaan Terutama Organ Ileum Yang Disebabkan Oleh Efek Pemberian NSAID Diantaranya Indometasin. Dosis Pemberian Indometasin 15mg/Kg BB Per Oral Dapat Menyebabkan IBD Pada Hewan Coba. Penelitian Ini Bertujuan Untuk Mengetahui Efek Terapi Perasan Buah Labu Siam (Sechium Edule) Terhadap Penurunan Aktivitas Protease Dan Perubahan Profil Protein Pada Ileum Tikus (Rattus Norvegicus). Hewan Coba Yang Dipakai Yaitu Tikus (Rattus Norvegicus) Jantan Berumur 8-12 Minggu Yang Dibagi Menjadi Empat Kelompok Yakni Kelompok 1 Yaitu Kontrol, Kelompok 2 Yaitu IBD, Kelompok 3 Dan 4 Yaitu Dosis 10g/Tikus Dan 20g/Tikus. Aktivitas Protease Diukur Menggunakan Metode Spektofotometri. Profil Protein Ileum Di Analisis Dengan SDS–PAGE. Hasil Penelitan Menunjukkan Bahwa Terapi Perasan Buah Labu Siam Secara Signifikan (P< 0,05) Menurunkan Aktivitas Protease. Dosis Efektif Terapi Adalah 20 G/Tikus Yang Menurunkan Aktivitas Protease Sebesar 46,95%. Hasil Analisis Profil Protein Menunjukkan Bahwa Protein BM 62 Kda Tidak Disintesis Setelah Terapi Perasan Buah Labu Siam Dan Protein Ini Diduga Sebagai Vasoactive Intestinal Polypeptide (VIP). Dapat Disimpulkan Bahwa Terapi Perasan Buah Labu Siam Dapat Menurunkan Aktivitas Protease Ileum Dan Menghambat Sintesis Protein 62 Kda.
English Abstract
Inflammatory Bowel Disease (IBD) Is A Digestive System Disease Especially In Ileum Organ Which Is Caused By The Effect Of NSAID Such As Indomethacine. The Dose Of 15 Mg/Kg BW Indomethacine Per Oral Resulted IBD On Rat Animal Model. The Research Was Aimed To Find Out The Effect Of Chayote (Sechium Edule) Extract Therapy Toward Protease Activity And Protein Profile Alteration In The Rats Ileum. The Research Used Male Rats (Rattus Norvegicus) Aged Of 8-12 Weeks Divided Into Four Groups. They Were Control Group, IBD Group, Therapy Group With Dose Of 10g/Rat And 20g/Rat Chayote Extract. Protease Activity Was Assessed By Spectrophotometry. Protein Profile Was Analyze Using SDS-PAGE. The Results Showed That Chayote Extract Therapy Decreased Protease Activity Significantly (P<0.05). The Effective Dose Was 20g/Rats, Decreased To Be 46.95% Of Protease Activity. The Protein Profile Analysis Showed That 62 Kda MW Assumed As Vasoactive Intestinal Polypeptide (VIP) Was Not Synthesized After Chayote Extract Therapy. In Conclusion, Chayote Extract Therapy Could Decrease Ileum Protease Activity And Prevent The Synthesize Of 62 Kda Protein.
