Abstract
Hiperkolesterolemia merupakan keadaan dimana kadar kolesterol khususnya Low Density Lipoprotein (LDL) dalam darah melebihi batas normal. LDL yang seharusnya menuju ke jaringan akan terakumulasi pada pembuluh darah yang nantinya akan menyebabkan oksidasi LDL pada pembuluh darah sehingga memicu terjadinya reaksi inflamasi. Tujuan penelitian ini adalah untuk mengetahui efek pencegahan yogurt susu kambing pada hewan model tikus (Rattus norvegicus) hiperkolesterolemia terhadap ekspresi Tumor Necrosis Factor Alpha (TNF- α) dan gambaran histopatologi pembuluh darah aorta. Penelitian ini menggunakan tikus (Rattus norvegicus) jantan, strain Wistar yang dibagi dalam 5 kelompok yaitu kelompok kontrol, kelompok hiperkolesterolemia, kelompok pencegahan dosis 300 mg/kg BB, dosis 600 mg/kg BB dan 900 mg/kg BB. Pencegahan yogurt susu kambing diberikan selama 28 hari kemudian dilanjutkan dengan pemberian diet hiperkolesterolemia bersamaan dengan pemberian yogurt selama 14 hari. Analisis yang digunakan dalam penelitian ini secara deskriptif kuantitatif ekspresi TNF- α pada jaringan pembuluh darah aorta dengan One Way Analisis Of Variance (ANOVA) apabila terdapat perbedaan yang nyata dilanjutkan uji Tukey 5% dan secara kualitatif dengan membandingkan gambaran histopatologi aorta dari masing-masing kelompok perlakuan. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang nyata (p<0,05) pada ekspresi TNF- α. Pencegahan dosis 900 mg/Kg BB merupakan dosis terbaik menurunkan ekspresi TNF-α. Selain itu yogurt susu kambing dapat mencegah kerusakan jaringan aorta yang ditunjukkan dengan keadaan lapisan aorta yang tersusun atas tunika intima, media dan adventisia mendekati keadaan tikus normal.
English Abstract
Hypercholesterolemia is a condition where the cholesterol levels specifically Low Density Lipoprotein (LDL) in the blood exceeds the normal limits. The increasing of LDL causes LDL oxidation in the vascular tissue that trigger the inflammatory reaction. The purpose of this study was to determine the preventive effect of goat milk yogurt in animal models of hypercholesterolemic rats (Rattus norvegicus) on the expression of Tumor Necrosis Factor Alpha (TNF- α) and aortic vascular histopathology. This study used male rats (Rattus norvegicus), strain Wistar were divided into 5 groups: control group, hypercholesterolemic group, prevention group was dose of 300 mg / kg BW, 600 mg / kg BW and 900 mg/kg BW. Prevention of hypercholesterolemia was conducted by administered yogurt for 28 days then gave dietary hypercholesterolemia with yogurt simultaneously for 14 days. The analysis used in this research was quantitative descriptive for expression of TNF - α in aortic with One Way Analysis of Variance (ANOVA) 5% and qualitatively by comparing the aortic histopathology from each group. The results showed that there were significantly difference (p <0.05) in the expression of TNF-α. Prevention dose of 900 mg / kg bw was the best dose to decrease expression of TNF-α. In addition, goat milk yogurt could prevent aortic tissue damage as indicated by the aorta was composed of layers of tunica intima, media and adventitia approaching a normal rat.
