Abstract
Hiperkolesterolemia merupakan suatu keadaan dimana terjadi peningkatan serum Low Density Lipoprotein (LDL) dalam tubuh diatas batas normal. Pemberian diet hiperkolesterol diketahui dapat menyebabkan keadaan hiperkolesterolemia. Yogurt susu kambing memiliki kandungan peptida bioaktif yang dapat menurunkan kadar kolesterol dalam darah. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian yogurt susu kambing terhadap ekspresi INOS dan gambaran histopatologi jaringan jantung. Tikus yang dipakai dalam penelitian ini adalah tikus putih (Rattus norvegicus) jantan berumur 12 minggu yang dibagi dalam 5 kelompok yaitu kelompok kontrol, kelompok hiperkolesterolemia, kelompok hiperkolesterolemia dengan terapi yogurt 300 mg/kg BB, kelompok hiperkolesterolemia dengan terapi yogurt 600 mg/kg BB, dan kelompok hiperkolesterolemia dengan terapi yogurt 900 mg/kg BB. Hasil dari penelitian ini menunjukkan bahwa pemberian terapi yogurt susu kambing memberikan pengaruh yang berbeda nyata (p<0,05) terhadap penurunan ekspresi INOS antar kelompok. Pada terapi dosis 900 mg/kg BB menunjukkan dosis terbaik dalam menurunkan ekspresi INOS yaitu 117%. Selain itu pemberian terapi yogurt susu kambing berpengaruh terhadap perbaikan histopatologi kardiomiosit yang ditunjukkan dengan struktur kardiomiosit tidak beraturan dan bergelombang menjadi lebih baik yaitu susunan serat miofibril menjadi teratur. Dapat disimpulkan bahwa terapi yogurt susu kambing dapat digunakan sebagai terapi alternatif dalam keadaan hiperkolesterolemia
English Abstract
Hypercholesterolemia is a condition of higher level of Low Density Lipoprotein serum (LDL) in the body. Giving hypercholesterol diet lead to hypercholesterolemia. Goat milk yogurt contents of bioactive peptide that can lower the cholesterol level in the blood. This research aimed to know the effect of giving goat milk yogurt towards the INOS expression and the heart histopathology. This research used male rats (Rattus norvegicus) aged 12 weeks that divided into 5 groups : control group, hypercholesterolemic group, hypercholesterolemic with 300 mg/kg BW goat milk yogurt therapy, hypercholesterolemic with 600 mg/kg BW goat milk yogurt therapy, and hypercholesterolemic with 900 mg/kg BW goat milk yogurt therapy. The results of this research indicated a decreasing of INOS expression significantly different between the treatments (p<0.05). The best deterioration was generated by the 900 mg/kg BW dose therapy. Morever, giving the goat milk yogurt therapy affected the improvement of the wavy cardiomyocytes became better with normal structure of myofibril. It could be concluded that the goat milk yogurt therapy could be used as alternative therapy for hypercholesterolemia.
