Abstract
Fibrosis ginjal merupakan akumulasi abnormal extracellular matrix (ECM) pada jaringan ginjal yang dapat merubah struktur ginjal normal. Fibrosis ginjal adalah manifestasi akhir dari Chronic Kidney Disease (CKD). Vitamin E merupakan antioksidan yang berperan merusak rantai radikal bebas yang merupakan inisiator terjadinya fibrosis ginjal sehingga berpotensi untuk dijadikan bahan terapi. Penelitian ini bertujuan untuk mengetahui potensi vitamin E sebagai terapi fibrosis ginjal pada hewan model tikus berdasarkan ekspresi iNOS dan gambaran histopatologi ginjal. Tikus model fibrosis ginjal disiapkan dengan injeksi Streptokinase secara intravena pada vena coccygea dengan dosis 6000IU/ekor tikus, streptokinase diinduksikan sebanyak tiga kali dengan rentang waktu lima hari. Penelitian dibagi menjadi 5 kelompok perlakuan, yaitu kelompok kontrol, kelompok fibrosis ginjal, kelompok fibrosis ginjal yang telah diterapi vitamin E dosis 200 mg/Kg BB, 300 mg/Kg BB, dan 400 mg/Kg BB. Ekspresi iNOS dihitung menggunakan software Axiovision dan dianalisis dengan ANOVA. Pengamatan gambaran histopatologi glomerulus ginjal menggunakan mikroskop dan dianalisis deskriptif. Hasil penelitian menunjukan pemberian vitamin E mampu menurunkan ekspresi iNOS secara signifikan (P<0,05) dan memperbaiki gambaran histopatologi glomerulus ginjal yang ditandai dengan perbaikan capsula bowman, dan tidak adanya glumerulosclerosis dan hemorraghi pada glomerulus. Dosis 400 mg/Kg BB adalah dosis vitamin E terbaik untuk terapi fibrosis ginjal. Kesimpulan dari penelitian ini adalah pemberian vitamin E dapat digunakan sebagai terapi pada tikus model fibrosis ginjal berdasarkan ekspresi iNOS dan gambaran histopatologi glomerulus ginjal.
English Abstract
Renal fibrosis is characterized by the abnormal accumulation of extracellular matrix (ECM) that affected normal kidney structures. Vitamin E is an antioxidant that plays role to damage free radical chain which the initiator of renal fibrosis. The purpose of this research was to study the potential of vitamin E affect to iNOS expression and renal glomerulus histopathology appearance on renal fibrosis rats. Renal fibrosis rats were prepared by intravenously injection of streptokinase with dose of 6000 IU, it was induced three times with five days interval. Five groups were used in this research were control group, renal fibrosis group, and three groups with therapy of vitamin E dose of 200 mg/Kg BW, 300 mg/Kg BW, and 400 mg/Kg BW. INOS expressions were determined by axiovision software and were analyzed by ANOVA. Histophatological observations of renal glomerulus were observed microscopicaly and were analyzed descriptively. The result showed that vitamin E therapy significantly (P<0.05) could decrease iNOS expressions more over it also repaired renal glomerulus damage in renal fibrosis rats were marked by repairing of capsula bowman and absence of glumerulosclerosis and hemorrahagi on glomerulus. The dose of 400 mg/Kg BW is the best vitamin E dose for renal fibrosis therapy. It can be concluded that vitamin E have possibility as therapy on renal fibrosis rats models based on iNOS expressions and renal glomerulus histopathology
