Abstract
Hiperkolesterolemia adalah suatu keadaan tingginya kadar kolesterol dalam darah yang disebabkan karena usia, makanan tinggi kolesterol dan asam lemak jenuh. Dilaporkan terjadi peningkatan kasus hiperkolesterolemia yang mengakibatkan penyakit kardiovaskular dan kematian tiap tahunnya. Yoghurt susu kambing mempunyai kandungan Bakteri Asam Laktat (BAL) dan antioksidan yang diduga mampu mencegah hiperkolesterolemia. Tujuan penelitian ini adalah untuk mengetahui pengaruh yoghurt susu kambing sebagai upaya pencegahan hiperkolesterolemia ditinjau dari kadar Malondialdehida (MDA) dan ekspresi inducible Nitric Oxide Synthase (iNOS). Penelitian ini menggunakan tikus putih strain Wistar dan dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif, kontrol positif hiperkolesterolemia, serta diet hiperkolesterol dengan pemberian yoghurt dosis 300, 600 dan 900 mg/kg BB. Yoghurt susu kambing diberikan selama 28 hari, dilanjutkan dengan pemberian yoghurt dan diet hiperkolesterol secara bersamaan selama 14 hari. Data yang diperoleh dianalisis dengan SPSS 16.0 dengan melakukan uji analisis varian (ANOVA) dan dilakukan analisis lanjutan dengan uji Tukey (α = 5 %). Hasil penelitian menunjukkan bahwa pencegahan hiperkolesterolemia dengan yoghurt susu kambing menurunkan kadar MDA secara signifikan (p<0,05) antar kelompok perlakuan, kelompok dengan dosis pemberian yoghurt 900 mg/Kg BB adalah dosis terbaik menurunkan kadar MDA dan ekspresi iNOS pada aorta. Kesimpulan penelitian ini adalah pencegahan hiperkolesterolemia dengan yoghurt susu kambing berpengaruh nyata menurunkan kadar MDA dan ekspresi iNOS
English Abstract
Hypercholesterolemia is a condition of high cholesterol levels in blood and have correlation with age, high foods cholesterol and saturated fatty acids. Increasing of hypercholesterolemic patients which caused cardiovascular disease and mortality has been reported each year. Goats milk yogurt known rich of Lactic Acid Bacteria (LAB) and antioxidants that are thought to be able to prevent hypercholesterolemia. The purpose of this study was to determine the effect of goat milk yogurt for the prevention of hypercholesterolemia based on Malondialdehyde (MDA) levels and the expression of inducible Nitric Oxide Synthase (iNOS) of aorta. This study used Rattus norvegicus Wistar strain which divided into 5 groups: negative control, hypercholesterolemia control (positive control), group of diet hypercholesterolemia with yogurt treatment doses of 300, 600, 900 mg / Kg BW. Goats milk yogurt was given for 28 days, followed with administration of yogurt and dietary hypercholesterolemia simultaneously for 14 days. Data were analyzed with analysis of variance (ANOVA) and further analyzed by Tukeys test (α = 5%). iNOS expression of aorta were also analyzed descriptively. The results showed that the MDA level was significantly different (P <0.05) between treatment groups. The preventive yogurt with dose of 900 mg / kg BW was the best dose that decreasing MDA level and iNOS expressions on aorta. It can be concluded that the prevention therapy of hypercholesterolemia with goat milk yogurt lowered both of MDA level and iNOS expression
