Abstract
Hiperkolesterolemia adalah suatu keadaan meningkatnya kadar kolesterol di dalam darah. Penyebab hiperkolesterolemia salah satunya adalah pemberian diet tinggi lemak yang menyebabkan peningkatan jumlah radikal bebas di dalam tubuh. Antioksidan endogen tidak mampu mengatasi radikal bebas yang berlebih sehingga diperlukan antioksidan yang berasal dari luar tubuh. Ekstrak air daun sukun (Artocarpus altilis) mengandung senyawa bersifat antioksidan yaitu flavonoid yang berupa kuersetin, DS-6, artoindonesianin F, dan cycloaltilisin menurunkan kadar trigliserida dan mengurangi perlemakan pada hepar. Tujuan penelitian ini adalah mengetahui pengaruh ekstrak air daun sukun terhadap penurunan kadar trigliserida dan perbaikan histopatologi hepar tikus (Rattus norvegicus) hiperkolesterolemia. Penelitian ini menggunakan hewan coba tikus yang dikelompokkan menjadi 5 yaitu kelompok kontrol negatif, kelompok kontrol positif, kelompok terapi dengan dosis 500 mg/Kg BB, 1000 mg/Kg BB dan 2000 mg/Kg BB. Pengujian kadar trigliserida menggunakan metode GPO-PAP. Analisa data kadar trigliserida menggunakan one way ANOVA α=0,05 dengan uji lanjutan uji Tukey, sedangkan histopatologi hepar secara kualitatif. Hasil penelitian menunjukkan terapi ekstrak air daun sukun menurunkan kadar trigliserida secara signifikan (p<0,05) antar perlakuan. Dosis terapi 2000 mg/kg BB efektif dalam menurunkan kadar trigliserida sebesar 32,89%. Disimpulkan bahwa ekstrak air daun sukun yang mengandung flavonoid dapat digunakan sebagai terapi hiperkolesterolemia berdasarkan penurunan kadar trigliserida dan perbaikan pada gambaran histopatologi hepar tikus.
English Abstract
Hypercholesterolemia is a condition where cholesterol level in blood serum increases. One of the causes of hypercholesterolemia is the consumption of high fatty diet which increases free radical substances in the body. Endogenous antioxidant in the body is not able to overcome excessive free radical substances, thus exogeneous antioxidant is needed. Breadfruit leaf (Artocarpus Altilis) has antioxidant compound namely flavonoid in the form of kuersetin DS-6, artoindonesianin F, and cycloaltilisin which can lower triglyceride level and repairing histopathology liver. The purpose of this research is to know the effects of breadfruit leaf water extract for lowering triglyceride level and improving hepar histopathology of hypercholesterolemic rats. This research uses rats as the experimental animals. The rats were divided into 5 groups: negative control group, positive control group, and therapy group with dose of 500 mg/kg bw, 1000 mg/kg bw, and 2000 mg/kg bw. One-way ANOVA α=0,05 with Tukey test is used in triglyceride level data analysis, whereas hepar histopathology is conducted with qualitative analysis. The result showed that breadfruit leaf water extract therapy lowers triglyceride level significantly (p<0,05) between groups. Therapeutic dose of 2000 mg/kg bw is the effective doses for lowering triglyceride level until 32,89%. In conclusion, breadfruit leaf water extract contaning flavonoid can be used as a treatment of hypercholesterolemia based on the lowering of triglyceride level and improvement of hepar histopathology on rats
