Abstract
Hiperkolesterolemia adalah keadaan konsentrasi kolesterol dalam darah melebihi batas normal. Kondisi hiperkolesterolemia memicu terjadinya inflamasi disebabkan radikal bebas dan peroksidasi lipid pada organ duodenum. Yoghurt susu kambing memiliki potensi sebagai antiinflamasi. Tujuan penelitian ini untuk mengetahui peran yoghurt susu kambing dalam mempertahankan ekspresi Inducible Nitric Oxide Synthase (iNOS) tikus (Rattus norvegicus) setelah diinduksi diet hiperkolesterol serta dalam mempertahankan gambaran histopatologi duodenum. Penelitian ini mengunakan hewan coba tikus (Rattus norvegicus) jantan strain Wistar yang dikelompokan menjadi 5 kelompok yaitu kelompok kontrol negatif, kelompok hiperkolesterolemia (kontrol positif), kelompok pencegahan yoghurt susu kambing dosis 300 mg/kg BB, 600 mg/kg BB dan 900 mg/kg BB. Starter yoghurt mengandung bakteri Lactobacillus bulgaricus, Lactobacillus acidophilus, dan Streptococcus thermophilus. Hewan model hiperkolesterolemia dibuat dengan pemberian diet hiperkolesterol mengandung minyak babi 10%, asam kholat 0,1% dan kuning telur puyuh rebus 5% . Tikus diberi yoghurt susu kambing selama 28 hari. Pada hari ke 36, tikus diberi diet hiperkolesterolemia beserta yoghurt susu kambing dengan metode sonde lambung selama 14 hari. Analisa data ekspresi iNOS menggunakan one way ANOVA dengan uji lanjutan uji Tukey α=0,05, sedangkan analisis histopatologi duodenum dilakukan secara kualitatif dengan metode pewarnaan Hemaktosilin–Eosin. Hasil penelitian menunjukkan bahwa pencegahan yoghurt susu kambing dapat mempertahankan ekspresi iNOS dan gambaran histopatologi duodenum. Ekspresi iNOS berbeda nyata antara perlakuan (p<0,05) dan dosis yoghurt yang paling signifikan adalah dosis yoghurt 900 mg/kg BB yang mampu mempertahankan ekspresi iNOS sebesar 46,51%. Gambaran histopatologi duodenum pada kelompok B menunjukkan adanya infiltrasi sel radang seperti makrofag, neutrofil, dan leukosit dan kerusakan epitel membran submukosa, sedangkan pada kelompok yang diberi pencegahan menunjukkan infiltrasi sel radang seperti makrofag, neutrofil, dan leukosit dan kerusakan epitel membran submukosa dalam jumlah yang lebih sedikit. Dari hasil penelitian ini dapat disimpulkan bahwa yoghurt susu kambing tidak mampu mempertahankan ekspresi iNOS dan kondisi normal histopatologi duodenum.
English Abstract
Hypercholesterolemia is condition which the consentration of cholesterol in a blood exceed normal condition. Hypercholesterolemia triggered inflammation due to free radicals and lipid peroxidation in duodenum. Goat milk yogurt had a potential as an anti-inflammation. The purpose of this study was to know the effect of goat milk yogurt in maintaining normal expression of Inducible Nitric Oxide Synthase (iNOS) of rat (Rattus norvegicus) after induced hypercholesterolemia and goat milk yogurt duodenal histopathology. This research used rats male Wistar strains were grouped into 5 groups: control group, hypercholesterolemia group, hypercholesterolemia group that received goat milk yogurt as preventive dose of 300 mg / kg, 600 mg / kg and 900 mg / kg BB. Yogurt starter contained the bacteria Lactobacillus bulgaricus, Lactobacillus acidophilus, and Streptococcus thermophilus. Animal model hypercholesteromic were fed goat milk yogurt by force feeding methode for 28 days. Then at 36 th day, they were administrated with hypercholesterolemia diet contained 10% lard, 0.1% cholic acid and 5% boiled quail egg yolks and yogurt simultaneously for 14 days. iNOS expression were analyzed used one way ANOVA with Tukey test α = 0.05, whereas duodenal histopathology analysis by qualitatively. The results showed that the prevention of goat milk yogurt could retain the iNOS expression. The iNOS expression in group B than in group A showed an increase, whereas the expression of iNOS in groups C, D, and E were given preventive yogurt showed that the expression approach the control group. The iNOS expression was significantly different between treatments (P <0,05) and the dose that showed the best result for preventing hypercholesterolemia was 900 mg / kg BW. Group B showed the presence of inflammatory cell infiltration and epithelial damaged submucosal membrane, whereas the preventive groups (C, D, and E) showed the reduction of inflammatory cell infiltration like macrofag, neutrofil, and leukosit and also epithelial damaged submucosal membrane. The result concluded that goat milk yogurt were potential to prevent the increasing INOS expression and damaging of duodenal histopathology
