Abstract
Merokok merupakan salah satu faktor yang berpengaruh dalam infertilitas. Hal ini dipicu adanya radikal bebas dalam asap rokok. Radikal bebas dalam tubuh dapat menyebabkan kerusakan DNA spermatozoa sehingga terjadi penurunan ekspresi protamine dan motilitas spermatozoa. Pemberian antioksidan berupa ekstrak etanol kulit buah manggis mampu mengikat radikal bebas yang ada dalam tubuh. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak etanol kulit buah manggis (Garcinia mangostana L) terhadap ekspresi protamine serta motilitas spermatozoa tikus (Rattus norvegicus) jantan yang terpapar asap rokok. Penelitian ini menggunakan tikus jantan berumur 3 bulan yang dibagi dalam 5 kelompok. Kelompok 1 adalah kelompok kontrol negatif (tikus sehat), kelompok 2 adalah kelompok kontrol positif yang diberikan paparan asap rokok, kelompok 3, 4 dan 5 adalah tikus yang dipapar asap rokok dan diberi terapi ekstrak etanol kulit buah manggis 200 mg/kgBB, 400 mg/kgBB dan 600 mg/kgBB. Ekspresi protamine diamati menggunakan metode imunohistokimia sedangkan pengamatan motilitas spermatozoa tikus diamati secara mikroskopis. Hasil penelitian menunjukkan terapi ekstrak etanol kulit buah manggis pada tikus jantan yang terpapar asap rokok mampu meningkatkan ekspresi protamine dan meningkatkan motilitas spermatozoa tikus secara nyata (p<0,05). Pemberian terapi ekstrak etanol kulit buah manggis dosis 600 mg/kgBB memberikan hasil terbaik dibuktikan dengan peningkatan ekspresi protamine sebesar 78.94% dan motilitas spermatozoa mendekati kontrol negatif.
English Abstract
Smoking is one of factors which influence infertility. It happens because smoke contains free radicals. Accumulation of free radicals in the body can affect the spermatozoa’s DNA damage, which lead to decrease the protamine expression and spermatozoa motility. Therapy of mangosteen peel’s ethanol extract could reduce free radical in the body. This research was aimed to find out the effect of mangosteen peel’s ethanol extract (Garcinia mangostana L) towards expression of protamine and spermatozoa motility of cigarette-smoked rat (Rattus norvegicus). This research used male-3-old-month-rats divided in 5 groups. The first group was negative control, the second group was cigarette-smoked rats as positive control, the third, fourth and fifth group ware cigarette-smoked rats given mangosteen peel ethanol extract therapy dose of 200mg/kgBW, 400mg/kgBW and 600mg/kgBW respectively. The expression of protamine was observed by using immunohistochemistry method while rat spermatozoa’s motility was observed microscopically. The result showed that mangosteen peel’s ethanol extract therapy could increase the expression of protamine and increase rat spermatozoa’s motility significantly (p<0.05). Therapy of mangosteen peel’s ethanol extract dose of 600 mg/kgBW showed the best result by the increasing of protamine expression up to 78.94% and spermatozoa’s motility as shown on negative control.
