Abstract
Autoimmune thyroiditis (AITD) merupakan penyakit autoimun yang menyebabkan gangguan fungsi fisiologis pada organ tiroid. Iodin merupakan salah satu faktor lingkungan yang berperan sebagai penyebab autoimun pada kelenjar tiroid. Yoghurt susu kambing diketahui mengandung peptida bioaktif yang memiliki efek sebagai antioksidan, antiinflamasi, dan imunomodulator. Penelitian ini bertujuan untuk mengetahui pengaruh terapi yoghurt susu kambing terhadap AITD melalui adanya penurunan kadar malondialdehide (MDA) dan perbaikan gambaran histopatologi pankreas tikus (Rattus norvegicus) model AITD yang diinduksi NaI 0,05% (m/v). Penelitian ini menggunakan hewan coba tikus (Rattus norvegicus) betina, umur 8-12 minggu, berat badan 100-150 gram. Hewan model AITD dibagi tiga kelompok, yaitu kelompok kontrol (A), kelompok perlakuan (B) suplementasi NaI 0,05% (m/v), dan kelompok terapi yoghurt susu kambing (C) dosis 109cfu/ml. Induksi AITD dilakukan dengan supplementasi NaI 0,05% (m/v) selama 5 minggu melalui air minum. Terapi yoghurt susu kambing dilakukan dengan dosis 109 cfu/ml sebanyak 1 ml selama 4 minggu secara per oral. Variabel yang diamati dalam penelitian ini adalah gambaran histopatologi kelenjar pankreas dengan pewarnaan HE dan kadar MDA pankreas diukur dengan metode Tiobarbituric Acid (TBA). Kadar MDA dianalisis secara statistik kuantitatif dan gambaran histopatologi diamati secara deskriptif kualitatif. Hasil penelitian menunjukkan terapi yoghurt susu kambing mampu menurunkan kadar MDA sebesar 51% pada kelompok terapi dibandingkan dengan kelompok AITD. Gambaran histopatologi menunjukkan adanya perbaikan kerusakan pankreas yang berupa perbaikan atrofi pulau Langerhans dan pengurangan jumlah sel nekrosis. Kesimpulan dari penelitian ini adalah yoghurt susu kambing mampu menurunkan kadar MDA dan mampu memperbaiki gambaran histopatologi kerusakan pankreas pada tikus model AITD.
English Abstract
Autoimmune thyroiditis (AITD) is an autoimmune disease that causes disruption to the physiological function of the thyroid organ. Iodine is one of environment factor which contribute to autoimmune causal in thyroid gland. Goats milk yogurt has been known containing peptide bioactive peptide that has an effect as an antioxidant, anti-inflammatory, and immunomodulator. This study was aimed to determine the therapy effect of goats milk yogurt to AITD through the decreasing the malondialdehyde (MDA) levels and the repairing of pancreatic histopathology on AITD rat models induced by 0.05% (m/v) NaI. This study used female rats, age 8-12 weeks, weight of 100-150 grams. Animal models of AITD divided into three groups: control group (A), treatment group (B) of 0.05% (m/v) NaI supplementation for, and goat milk yogurt therapy group (C) dose of 109 cfu/ml. Induction of AITD was performed with 0.05% (m/v) NaI supplementation for 5 weeks through the drinking water. Goat milk yogurt therapy was done with a dose of 109 cfu/ml as 1 ml for 4 weeks in per oral. The variables observed in this study was the histopathology of the pancreas gland with HE staining and pancreatic MDA levels that were measured by Thiobarbituric Acid (TBA) method. MDA levels were analyzed quantitatively and histopathology were analyzed qualitatively. The results showed the goats milk yogurt therapy could reduce MDA levels to 51% in therapy group compared with AITD group. Histopathology showed repairement of pancreatic damage in the form of atrophy of the islets of Langerhans’s and the a reduction in the amount of necrosis cells. The conclusion of this study was the goats milk yogurt could reduce MDA levels and able to repair the damaging of pancreas histopathology in rat models of AITD.
