Abstract
Asap rokok merupakan salah satu sumber radikal bebas eksogen yang dapat melemahkan pertahanan antioksidan sehingga kadar radikal bebas dalam tubuh meningkat. Peningkatan radikal bebas dapat mempengaruhi organ reproduksi yang menyebabkan penurunan kualitas dan kuantitas sperma. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian ekstrak kulit buah manggis (Garcinia mangostana L.) yang mengandung antioksidan terhadap kadar malondialdehyde (MDA) pada organ testis dan jumlah spermatozoa. Penelitian ini menggunakan tikus (Rattus norvegicus) jantan berumur 3 bulan yang dibagi dalam 5 kelompok yaitu kelompok kontrol negatif (tidak dipapar asap rokok), kelompok kontrol positif (dipapar asap rokok), kelompok dipapar asap rokok dengan terapi ekstrak kulit buah manggis dosis 200 mg/kgBB, dosis 400 mg/kgBB, dan dosis 600 mg/kgBB. Paparan asap rokok diberikan 2 batang/hari/kelompok dengan cara memasukkan asap rokok ke dalam bak kedap udara selama 30 hari. Terapi ekstrak kulit buah manggis dilakukan dengan cara per oral menggunakan sonde selama 21 hari. Kadar MDA diukur dengan metode Thiobarbituric Acid (TBA) dan penghitungan jumlah spermatozoa dengan menggunakan alat hemocytometer. Hasil penelitian menunjukkan bahwa terapi ekstrak kulit buah manggis dosis 200, 400 dan 600 mg/kgBB pada tikus yang dipapar asap rokok secara signifikan (p < 0,05) menurunkan kadar MDA sampai 53% dan meningkatkan jumlah spermatozoa sampai 71%. Kesimpulan dari penelitian ini adalah bahwa ekstrak kulit buah manggis mengandung antioksidan yang mampu menurunkan kadar MDA dan meningkatkan jumlah spermatozoa tikus yang terpapar asap rokok.
English Abstract
Cigarette smoke is one of the potential of exogenous free radicals which can weaken antioxidant system so that can increase free radicals levels in the body. The increasing of free radicals can affect reproduction organs causing the decrease of sperms’ quality and quantity. This study aimed to examine the effect of Mangosteen peel (Garcinia mangostana L.) extract which contains antioxidants to the malondialdehyde’s (MDA) levels in testis organ and the amount of sperm. This research used male-3-old-month-rats divided in 5 groups, they were negative control group, positive control group (cigarette smoked rats), cigarette-smoked rats given mangosteen peel extract therapy 200 mg/kgBW, 400 mg/kgBW, and 600 mg/kgBW. Cigarette smoke inducted by putting the smoke of 2 cigarettes in a vacum box per day per group for 30 days. The therapy conducted per-oral by using sonde for 21 days. The MDA levels were measured using Thiobarbituric acid (TBA) method and the amount of sperm calculated using a hemocytometer. The result of the research showed that the effect of mangosteen peel extract therapy 200, 400 and 600 mg/kgBW toward cigarette smoked rats significantly (p < 0.05) decreased the MDA levels up to 53 % and increased the amount of spermatozoa up to 71 %. It can be concluded that mangosteen peel extract contains antioxidant which is able to decrease MDA levels and increase the amount of cigarette smoked rat’s spermatozoa.
