Abstract
Inflammatory Bowel Disease (IBD) merupakan penyakit inflamasi pada saluran pencernaan. Inflammatory Bowel Disease (IBD) disebabkan oleh efek samping indometasin. Indometasin diberikan secara oral dengan dosis 15 mg/kg BB yang dapat menyebabkan inflamasi akut pada jejunum. Penelitian ini bertujuan untuk mengetahui efek terapi perasan buah labu siam (Sechium edule) dalam menurunkan aktivitas protease dan ekspresi TNF-α pada jejunum tikus (Rattus norvegicus) setelah mendapat paparan indometasin. Tikus (Rattus norvegicus) jantan strain Wistar berumur 8-12 minggu dengan berat 150-200 gram dibagi menjadi 4 perlakuan, yaitu tikus kontrol, tikus yang diinduksi indometasin, tikus yang diinduksi indometasin dan diberi terapi perasan labu siam (Sechium edule) sebanyak 10 gram/ekor, dan tikus yang diinduksi indometasin dan diberi terapi perasan labu siam (Sechium edule) sebanyak 20 gram/ekor. Aktivitas protease diukur dengan menggunakan metode spektrofotometri dan ekspresi TNF-α dengan teknik imunohistokimia. Hasil penelitian menunjukkan bahwa terapi perasan buah labu siam (Sechium edule) 10 gram/ekor dan 20 gram/ekor secara signifikan (p< 0,05) mampu menurunkan aktivitas protease dan ekspresi TNF- α. Nilai aktivitas protease pada kelompok terapi perasan buah labu siam 10 gram/ekor dan 20 gram/ekor yaitu 0,1462 ± 0,007588 μmol/ml.menit dan 0,1109 ± 0,001023 μmol/ml.menit. Ekspresi TNF-α kelompok terapi perasan buah labu siam 10 gram/ekor yaitu 1,7369 ± 0,3105 dan kelompok terapi perasan buah labu siam 20 gram/ekor yaitu1,1947 ± 0,2535. Kesimpulan dari penelitian ini menunjukkan bahwa terapi perasan buah labu siam (Sechium edule) dari 20 gram/ekor menurunkan aktivitas protease dan ekspresi TNF-α berturut-turut 44 % dan 61 %.
English Abstract
Inflammatory Bowel Disease (IBD) is a digestive inflammatory disease caused by the side effect of indomethacin. 15 mg/kg BW indomethacin was given orally to cause severe inflammation in jejunum. The purpose of this study is to describe the effect of chayotte (Sechium edule) squeeze therapy in decreasing protease activity and TNF- α in rat (Rattus norvegicus) jejunum after being exposed to indomethacin. In this research, 8-12- week male Wistar strain rats with 150-200 g body weight were divided into four experimental groups, those were controll-rats, indomethacin induction rats, indomethacin induction rats treated with chayotte (Sechium edule) squeeze therapy 10 gram/rat, and indomethacin induction rats treated with chayotte (Sechium edule) squeeze therapy 20 gram/rat. Protease activity was measured by spectrophotometry method, whereas TNF- α was examined by immunohistochemistry method. The experiment result concludes that chayotte (Sechium edule) squeeze therapy 10 gram/rat or 20 gram/rat can decrease protease activity and TNF-α expression significantly (p<0.05). The level of protease activity of 10 gram/rat and 20 gram/rat chayotte (Sechium edule) squeeze therapy group are 0.1462 ± 0.007588 μmol/ ml.minutes and 0,1109 ± 0,001023 μmol/ml.minutes. The TNF-α expression for 10 gram/rat chayotte (Sechium edule) squeeze therapy group is 1.7369 ± 0.3105 and for the 20 gram/rat chayotte (Sechium edule) squeeze therapy group is 1947 ± 0.2535. It can be concluded that chayotte (Sechium edule) squeeze therapy 20 gram/rat decreases protease activity and TNF-α expression of 44 % and 61 % respectively.
