Abstract

Fibrosis ginjal merupakan kelainan yang disebabkan oleh sekresi matriks ekstraselular berlebihan yang dapat terjadi pada semua penyakit ginjal kronik. Fibrosis ginjal dapat dipicu penggunaan Streptokinase. Tujuan penelitian ini adalah untuk mengetahui penurunan ekspresi TGF- dan perbaikan gambaran histopatologi ginjal pada tikus (Rattus norvegicus) fibrosis ginjal yang diinduksi streptokinase pasca terapi air rebusan kacang kedelai. Pada penelitian ini terdapat empat kelompok perlakuan yaitu kontrol (A), fibrosis ginjal (B), fibrosis ginjal yang diberi terapi air rebusan kacang kedelai dosis 6 g/2 ml/ekor (C), dan fibrosis ginjal yang diberi terapi air rebusan kacang kedelai dosis 9 g/2 ml/ekor (D). Parameter yang diamati dalam penelitian ini adalah ekspresi TGF- yang diamati dengan metode imunohistokimia dan gambaran histopatologi ginjal dengan pewarnaan Hematoxylen-Eosin. Hasil penelitian menunjukkan bahwa terapi air rebusan kacang kedelai secara signifikan (P<0,05) menurunkan ekspresi TGF-β. Penurunan ekspresi TGF-β terbaik dihasilkan pada kelompok fibrosis ginjal yang diberi terapi air rebusan kacang kedelai dosis 9 g/2 ml/ekor yaitu sebesar 71% sedangkan pada kelompok fibrosis ginjal yang diberi terapi air rebusan kacang kedelai dosis 6 g/2 ml/ekor terjadi penurunan sebesar 59%. Terapi air rebusan kacang kedelai juga menunjukkan adanya perbaikan histopatologi ginjal yang ditandai dengan penurunan akumulasi myofibroblas serta penurunan kerusakan epitel tubulus. Perbaikan gambaran histopatologi ginjal yang terbaik terjadi pada kelompok fibrosis ginjal yang diberi terapi air rebusan kacang kedelai dosis 9 g/2 ml/ekor.

English Abstract

Renal fibrosis is a disorder caused by excessive secretion of extracelullar matrix than can occur in all chronic kidney disease. One of the triggers that cause renal fibrosis is the usage of Streptokinase. The purpose of this study was to determine the decreasing of TGF-β expression and the improvement of kidney histopathology in rats (Rattus norvegicus) renal fibrosis induced by streptokinase after soybean stew water therapy. In this study, there were four treatment groups, were: control (A), renal fibrosis (B), renal fibrosis with soybean stew water therapy dose of 6 g/2 ml/rats (C), and renal fibrosis with soybean stew water therapy dose of 9 g/2 ml/rats (D). Parameters observed in this study were the TGF-β expression observed by immunohistochemistry methods and an appearance of renal histopathology with Hematoxylen-Eosin staining. The result of this study showed that soybean stew water therapy was significantly reduced (P<0,05) the expression of TGF-β. The lowest TGF-β expression resulted in soybean stew water therapy dose of 9 g/2 ml/rats. Soybean stew water therapy showed an improvement of kidney histopathology that characterized by decreasing of myofibroblast amount and decreasing in epithelial tubular damaged, the best improvement occured at soybean stew water therapy dose of 9 g/2 ml/rats.
