Abstract

Hipertensi merupakan peningkatan tekanan darah yang ditandai dengan tekanan darah sistolik lebih besar dari 140 mmHg dan tekanan diastolik lebih besar dari 90 mmHg. Induksi Deoxycorticosterone Acetate (DOCA) merupakan salah satu cara yang digunakan untuk membuat hewan model hipertensi. Induksi DOCA-salt dapat menyebabkan peningkatan Reactive Oxygen Species (ROS), terbentuk reaksi peroksidasi lipid sehingga terjadi peningkatan MDA. Yogurt susu kambing mengandung peptida bioaktif yang memiliki efek antihipertensi dan antioksidan. Tujuan dari penelitian ini untuk mengetahui pengaruh terapi yogurt susu kambing terhadap kadar MDA dan histopatologi ginjal tikus (Rattus norvegicus) model hipertensi yang diinduksi DOCA-salt. Penelitian ini dibagi dalam 5 kelompok yaitu kelompok kontrol negatif, kelompok hipertensi, terapi captopril, terapi yogurt dosis 300 mg/kg BB dan terapi yogurt dosis 600 mg/kg BB. Hasil pengukuran kadar MDA menunjukkan bahwa pemberian terapi yogurt dosis 300 mg/kg BB (0,471±0,023 μg/mL) dan yogurt dosis 600 mg/kg BB (0,406±0,025 μg/mL) dapat menurunkan kadar MDA lebih efektif daripada captopril (0,528±0,075 μg/mL). Kadar MDA kelompok terapi apabila dibandingkan dengan kelompok negatif yang paling efektif adalah kelompok yogurt dosis 600mg/kg BB. Hasil pengamatan histopatologi ginjal menunjukkan bahwa pemberian terapi yogurt susu kambing dan captopril dapat memperbaiki kerusakan glomerulus dan nekrosis tubulus. Kesimpulan dari penelitian ini adalah terapi yogurt susu kambing dapat menurunkan kadar MDA dan memperbaiki histopatologi ginjal tikus model hipertensi hasil induksi DOCA-salt.

English Abstract

Hypertension is a marked increase in blood pressure when the systolic blood pressure is greater than 140 mmHg and a diastolic pressure is greater than 90 mmHg. Deoxycorticosterone acetate (DOCA) is an agent commonly used to induce hypertension in experimental rats. DOCA-salt induction elevates reactive oxygen species (ROS) causing lipid peroxidation reaction indicated by increasing of MDA level. Goat milk yogurt contains bioactive peptide that has anti-hypertensive and anti-oxidant effects. This research was aimed to evaluate the therapeutic effects of goat milk yogurt to MDA level and kidney histopathology of hypertension rats model induced DOCA-salt. There were 2 control groups consisted of negative control and positive control, and 3 treated groups which were groups treated with captopril, 300 mg/kg body weight of yogurt, or 600 mg/kg body weight of yogurt. The measurement results indicated that MDA levels 300 mg/kg body weight of yogurt and 600 mg/kg body weight of goat milk yogurt therapy were more effective to reduce MDA level (0.471±0.023 μg/mL and 0.406±0.025 μg/mL) than captopril therapy (0.528±0.075 μg/ml). MDA level treatment group compared with the negative control is a group of the most effective 600 mg/kg body weight of goat milk yogurt. Histopatologic observation of kidney showed that the treatment of both goat milk yogurt and captopril could repair gromelurus damage and tubulus necrosis. Its proved that goat milk yogurt therapy could reduce MDA level and repair kidney histopathology of hypertension rats model induced DOCA-salt.
