Abstract
Autoimmune thyroiditis (AITD) merupakan penyakit autoimun yang menyebabkan kerusakan jaringan atau gangguan fungsi fisiologis pada organ tiroid. Pembuatan hewan model AITD bisa dilakukan dengan pemberian inducer seperti tiroglobulin (Tg). Dampak AITD hasil induksi Capra hircus thyroglobulin (cTg) terhadap kerusakan pankreas masih belum banyak dipelajari. Penelitian ini bertujuan untuk mengetahui kerusakan pankreas dengan adanya peningkatan kadar malondialdehid (MDA) dan gambaran histopatologi pankreas tikus ( Rattus norvegicus ) setelah diinduksi cTg. Hewan model AITD dalam penelitian ini dibuat dengan injeksi emulsi cTg dan Complete Freud’s Adjuvant (CFA) atau Incomplete Freud’s Adjuvant (IFA) (perbandingan 1 : 1) sebanyak 0,2 ml secara subkutan cervical pada hari ke-0, 14 dan 28. Nilai kadar MDA menunjukkan perbedaan yang nyata pada kelompok induksi cTg dosis 200 μg/mL dibandingkan dengan kontrol (p<0,05). Peningkatan kadar MDA sebesar 63% pada dosis 200 μg/mL dan 29% pada dosis 100 μg/mL dibandingkan dengan kelompok kontrol. Gambaran histopatologi menunjukkan adanya peningkatan kerusakan pankreas tikus berupa kariolisis dan atrofi dengan semakin bertambahnya dosis cTg. Kesimpulan dari penelitian ini adalah induksi cTg mampu meningkatkan kadar MDA dan menyebabkan perubahan gambaran histopatologi pankreas tikus.
English Abstract
Autoimmune thyroiditis (AITD) is an autoimmune disease which caused tissue damage or disruption to the physiological function of the thyroid organ. Preparation of animal models of AITD can be administered an inducer such as thyroglobulin (Tg) to rats. The impact of AITD induced by Capra hircus thyroglobulin (cTg) on pancreatic damage was not widely studied. This research was aimed to determine the pancreatic damage by the levels of malondialdehyde (MDA) and pancreas histopathology of rat ( Rattus norvegicus ) after induced by cTg. Animal models of AITD in this research were prepared by injecting emulsion of cTg and Complete Freud s Adjuvant (CFA) or Incomplete Freud s Adjuvant (IFA) (ratio 1:1) as much as 0.2 ml subcutaneously in the cervical, on days 0, 14 and 28. The MDA levels showed that there were significant differences in the groups cTg induction dose of 200 μg/mL compared to control (p<0.05). Increased MDA levels were 63% at a dose of 200 μg/mL and 29% at a dose of 100 μg/mL compared with the control group. Histopathology showed that there was increased in rat pancreatic damage by forming karyolysis and atrophy with the increasing of cTg dose. The conclusion of this research was the MDA levels can be increased by induction cTg and caused damage in histopathology of rat pancreas.
