Abstract
Arthritis Rheumatoid merupakan salah satu jenis penyakit autoimun yang menyerang bagian sendi. Penyebab arthritis rheumatoid meliputi faktor genetik, hormon sex, infeksi dan umur. Pengobatan yang biasa digunakan yaitu dengan beraktivitas ringan dan obat sintesis berbahan herbal yang mengandung antioksidan khususnya golongan polifenol. Tujuan penelitian ini yaitu untuk mengetahui ekspresi TNF-α dan gambaran histopatologi sendi pada tikus artritis rheumatoid yang telah diterapi dengan ekstrak buah kesemek junggo. Dalam penelitian ini, tikus dibagi dalam 4 kelompok yaitu kontrol (A), arthritis rheumatoid (B), arthritis rheumatoid dengan terapi ekstrak buah kesemek junggo 750 mg/kgBB (C) dan arthritis rheumatoid dengan terapi ekstrak buah kesemek junggo 1000 mg/kgBB (D). Induksi arthritis rheumatoid dilakukan dengan injeksi dengan Complete Freund Adjuvant (CFA) sebanyak 0.1 ml pada bagian ekor. Booster dilakukan setelah 1 minggu pada bagian kaki belakang masing-masing 0,05 ml per ekor. Terapi dilakukan selama 2 minggu sebanyak 2 ml/ekor/hari setelah injeksi ke II. Hasil dari penelitian ini menunjukkan bahwa tulang rawan hialin pada tikus arthritis rheumatoid mengalami kerusakan. Setelah dilakukan terapi (kelompok C dan D) terjadi perbaikkan struktur tulang rawan hialin. Ekspresi TNF-α ditunjukkan dengan persen area yaitu kelompok kontrol 0%, arthritis rheumatoid 475%, arthritis rheumatoid dengan terapi ekstrak kesemek junggo 750 mg/kgBB 62% dan arthritis rheumatoid dengan terapi ekstrak kesemek junggo 1000 mg/kgBB 44%. Hasil tersebut menunjukkan bahwa perlakuan terapi ekstrak buah kesemek junggo pada kelompok (C) dan (D) memiliki ekspresi TNF-α yang jauh lebih rendah bila dibandingkan pada tikus arthritis rheumatoid (B).
English Abstract
Rheumatoid arthritis is a type of autoimmune disease that attacks the joints. The etiology of rheumatoid arthritis include genetic factors, sex hormones, infection and ageing. The common therapy rheumatoid arthritis is by physical therapy and by administering herbal drug with antioxidant activity such as poliphenol. The purpose of this study was to determine the expression of TNF-α and histopathologic apperance of rheumatoid arthritis joints in rat that had been treated with an extract of persimmon junggo. In this study, rats were divided into 4 groups: control (A), rheumatoid arthritis (B), rheumatoid arthritis with junggo persimmon therapy of 750 mg/kg BW (C) and rheumatoid arthritis with jumggo persimmon therapy of 1000 mg/kg BW (D). Induction of rheumatoid arthritis was by conducted injection with Complete Freund Adjuvant (CFA) 0.1 ml on the tail. Booster made after 1 week on the back side of leg 0.05 ml/rat. Junggo persimmon ekstrack therapy wen conducted for 2 weeks as 2 ml/rat/day. Results from this eksperimen indicated that the hyaline cartilage in rat rheumatoid arthritis here damaged. After therapy (group C and D) show at that hyaline cartilage was repaired. Expression of TNF-α was shown by the percent area of were 0%, 475%, 62% and 44% fil group A, B, C and D recpectively. It could be concluded that therapy of junggo persimmon extract could decrease the expression of TNF-α in rat rheumatoid arthritis.
