Abstract

Jaminan keamanan pangan produk asal hewan terutama daging ayam di Indonesia perlu ditingkatkan untuk menghindari foodborne disease. Salah satu bakteri patogen yang sering ditemukan pada daging ayam adalah Salmonella. Cemaran Salmonella pada daging ayam salah satunya dapat disebabkan oleh kondisi di lingkungan RPA. Penelitian ini bertujuan untuk mengetahui adanya cemaran dan keragaman Salmonella spp., di lingkungan RPA. Sampel yang diteliti meliputi bulu ayam pasca pemotongan, air limbah cucian karkas, dan air minum ayam. Isolasi dan karakterisasi dilakukan berdasarkan sifat fenotip serta profil pita protein menggunakan metode SDS-PAGE. Hasil penelitian menunjukkan adanya cemaran Salmonella spp. yang tinggi yaitu lebih dari 106 cfu/ml. Pada penanaman di media XLD secara duplo dihasilkan 96 koloni dugaan Salmonella. Berdasarkan kemampuan tumbuh dan pengamatan morfologi koloni dihasilkan 25 isolat dugaan Salmonella, kemudian dilanjutkan uji biokimia dan uji serologi, dari uji tersebut dihasilkan 9 isolat yang menunjukan karakter Salmonella spp. Analisis pita protein dengan metode SDS-PAGE dapat terdeteksi pita protein dengan berat molekul 20,19 kDa, merupakan protein SefA yang dimiliki Salmonella enteriditis yaitu isolat L2A1, dan D1A2, pita protein dengan berat molekul 36,3 kDa yang merupakan protein adhesion fimbrae yang dimiliki Salmonella typhimurium yaitu isolat L2C2,B2A2, L1A2, dan L1B2, dan pita protein dengan berat molekul 25 kDa merupakan Salmonella arizonae adalah isolat B2A1, D2C2, dan D1C2. Kesimpulan yang dapat diambil dari penelitian ini yaitu terdapat cemaran Salmonella spp. dan keragaman Salmonella spp. di lingkungan RPA.

English Abstract

Food safety assurance from animal such as broilers in Indonesia needs increased to prevent foodborne disease. One of the common bacterial pathogens are Salmonella spp. in carcasses. Salmonella spp. contamination in carcasses can caused by slaughterhouse environment. This study was aimed to discover distribution and diversity of Salmonella spp. from samples in slaughterhouse. Samples taken from feathers after slaughtering, wasted water, and drink water. Isolation and characterization is started based on phenotype character of protein profile using SDS-PAGE method. This study showed that contamination of Salmonella spp. more than 106 cfu/ml. Bacterial growth in Xylose Lysine Deoxycholate Agar (XLD) has resulted 96 colony assumptive of Salmonella spp, after morphology observation resulted 25 assumptive Salmonella spp. The process had been continued by biochemistry test and serology test, resulted 9 isolates shown as Salmonella spp. Analized of protein profile using SDS-PAGE showed that band with 20.19 kDa weight molecules was SefA protein from Salmonella enteriditis were isolates L2A1, and D1A2, and band with 36.3 kDa weight molecules was adhesion fimbrae protein of Salmonella typhimurium were found in L2C2, B2A2, L1A2, and L1B2. The last, 25 kDa of weight molecules protein to be Salmonella arizonae were found in B2A1, D2C2 and D1C2. This study concluded that there were Salmonella spp. of contamination and variety of Salmonella taken from poultry slaughterhouse.
