Abstract

Tebu Saccharum officinarum L. merupakan tanaman perkebunan yang memiliki peran penting karena digunakan sebagai bahan baku untuk industri pangan dan farmasi. Saat ini terdapat teknologi pembibitan tebu yang lebih mudah dan efisien yakni dengan menggunakan satu mata tunas Single Bud Set. Pemanfaatan jamur rhizosfer sebagai pemacu pertumbuhan tanaman biasa dikenal sebagai PGPF pada pembibitan tanaman tebu belum banyak diketahui, sehingga penelitian mengenai jenis jamur yang berpotensi sebagai PGPF perlu dikaji. Pengetahuan mengenai jenis jamur rhizosfer sebagai PGPF akan memberikan informasi dalam pembibitan single bud set tanaman tebu. Penelitian dilaksanakan pada bulan Februari hingga Juli di Laboratorium Penyakit Tumbuhan, Jurusan Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Brawijaya, Malang dan Desa Sempalwadak, Kec. Bululawang, Kab. Malang. Penelitian seleksi jamur rhizosfer menggunakan benih mentimun terdiri dari 10 perlakuan dan 3 ulangan, dengan metode rancangan acak lengkap (RAL). Penelitian uji potensi jamur sebagai PGPF pada bibit single bud set tanaman tebu menggunakan metode Rancangan Acak kelompok (RAK) terdiri dari 8 perlakuan dan 3 ulangan, meliputi P0 (Perendaman bibit tebu dengan air), PK (Perendaman bibit tebu secara Konvensional), PR (Perendaman bibit tebu dengan PGPR Bacillus subtilis), PF1 (Perendaman Bibit tebu dengan isolat jamur Penicillium sp. isolat 1, PF2 (Perendaman Bibit tebu dengan isolat jamur Penicillium sp. isolat 2), PF3 (Perendaman Bibit tebu dengan isolat jamur Acremonium sp.), PF4 (Perendaman Bibit tebu dengan isolat jamur Trichoderma sp. isolat 1), dan PF5 (Perendaman Bibit tebu dengan isolat jamur Trichoderma sp. isolat 2). Data hasil pengamatan dianalisis dengan menggunakan analisis ragam (ANOVA), dan uji lanjut Duncan dengan taraf kesalahan 5%. Hasil seleksi jamur rhizosfer pada tanaman mentimun menunjukkan bahwa Acremonium sp, Penicillium spp. dan Trichoderma spp. berpotensi sebagai PGPF. Hasil penelitian uji potensi jamur menunjukkan bahwa tidak ada perbedaan yang nyata pada pengamatan yang meliputi presentase tunas tumbuh, jumlah anakan, serta kejadian penyakit, dan menunjukkan perbedaan yang nyata pada pengamatan tinggi tanaman, diameter batang dan jumlah daun tanaman tebu. Perendaman bibit single bud set dengan jamur Penicillium sp. isolat 2 berpotensi untuk memacu pertumbuhan tanaman tebu khususnya pada waktu tumbuh tunas tebu, tinggi tanaman, jumlah daun serta diameter batang. Perendaman bibit single bud set tanaman tebu dengan menggunakan jamur Trichoderma sp. isolat 1 dan isolat 2 berpotensi untuk mengurangi peluang kejadian penyakit.

English Abstract

Sugarcane (Saccharum officinarum L.) is a crop that has an important role because it is used as raw materials for food and pharmaceutical industries. Currently, there is more efficient sugarcane breeding technology using single bud bud set. There is not many reports on the utilization of rhizosphere fungal as PGPF in sugarcane planting, so research on the types of fungi that has potential as PGPF needs to be studied. Knowledge of the rhizosphere fungal species as PGPF will provide information in seeding single bud sugarcane set. The research was conducted from February to July in Plant Disease Laboratory, Department of Pest and Plant Disease, Agriculture Faculty, University of Brawijaya, Malang and Sempalwadak Village, Bululawang, Malang. Research on rhizosphere fungal selection using cucumber seeds consisted of ten treatments and three replications, with a Completely Randomized Design Method. Research on potential of rhizosphere fungal as PGPF on single bud sugarcane set with a Randomized Block Design consisted of eight treatments and three replicates, including P0 (Soaking sugarcane seedlings with water), PK (Soaking sugarcane seeds conventionally), PR (Soaking sugarcane seeds with PGPR Bacillus subtilis), PF1 (Soaking sugarcane seeds with Penicillium sp. isolate 1), PF2 (Soaking sugarcane seeds with Penicillium sp. isolate 2), PF3 (Soaking sugarcane seeds with Acremonium sp.), PF 4 (Soaking sugarcane seeds with Trichoderma sp. isolates 1, and PF5 (Soaking sugarcane seeds with Trichoderma sp. isolates 2). Observation data were analyzed using variance analysis (ANOVA), and Duncan's further test with α=5% error level. The results of the rhizosphere fungal selection using cucumber seeds showed that Acremonium sp, Penicillium spp. and Trichoderma spp. potential as PGPF. The results of the potential of rhizosphere fungal as PGPF on single bud sugarcane set showed that there were no significant differences in observations which included the percentage of germinating, number of tillers, and incidence of disease, and showed significant differences in observations of germinating time, plant height, stem diameter and number of sugarcane leaves. Soaking single bud sugarcane set with Penicillium sp. isolate 2 has the potential to increase the growth of sugar cane, especially germinating time, plant height, number of leaves and stem diameter. Soaking the single bud sugarcane set with Trichoderma sp. isolate 1 and isolate 2 has the potential to decrease the desease incidence.
