Abstract

Karies gigi merupakan proses demineralisasi yang menyerang jaringan keras gigi disebabkan oleh aktivitas mikroba yang meragikan karbohidrat Streptococcus mutans merupakan salah satu bakteri yang bereperan penting dalam karies gigi. Kulit apel malang dan apel washington merupakan salah satu bahan alami yang dapat berfungsi sebagai antibakteri karena diduga mengandung bahan aktif seperti flavonoid dan fenol. Penelitian ini bertujuan untuk membuktikan terdapat perbedaan efektivitas antibakteri antara ekstrak kulit apel malang dan apel washington terhadap bakteri Streptococcus mutans secara in vitro. Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan metode dilusi tabung untuk menentukan Kadar Hambat Minimum (KHM) dan Kadar Bunuh Minimum (KBM). Sampel yang digunakan dalam penelitian ini adalah isolat Streptococcus mutans yang diperoleh dari Laboratorium Mikrobiologi, Fakultas Kedokteran Universitas Brawijaya, Malang. Konsentrasi ekstak yang digunakan dalam penelitian, yaitu 0%, 1%, 2%, 3%, 4%, dan 5% v/v dengan empat kali pengulangan. Konsentrasi Streptococcus mutans sebesar 106CFU/ml. Hasil uji statistik menunjukkan bahwa kelompok ekstrak etanol kulit apel malang dan apel washington memiliki perbedaan yang tidak signifikan (Two Way Anova, p>0.05), namun terdapat perbedaan signifikan dari setiap pasangan kelompok konsentrasi (Post Hoc Tukey, p<0,05). KHM kedua ekstrak etanol kulit apel malang dan apel washington terhadap Streptococcus mutans adalah 3%. KBM ekstrak etanol kulit apel malang adalah 4% dan apel washington 5%. Dengan demikian, dapat disimpulkan bahwa terdapat perbedaan efektivitas antibakteri ekstrak etanol kulit apel malang dengan apel washington terhadap bakteri Streptococcus mutans secara in vitro namun tidak signifikan.

English Abstract

Dental caries is a demineralization process attacking dental hard tissue caused by microbial activities leaven the carbohydrates. Streptococcus mutans is one of the bacterium that plays an important role in dental caries. Rome beauty apple peels and washington apple peels are natural substances that can perform as an antibacterial because its suspected to contain active substances such as flavonoids and phenols. The experiments aimed to understanding the differentiation of the effectivity between ethanolic extract of rome beauty apple peel and washington apple peel towards Streptococcus mutans. The experiment of laboratoric experimental is used tube dilution test to determine the Minimum Inhibitory Concentration (MIC) and to determine the Minimum Bacterycidal Concentration (MBC). The sample used in this experiment was Streptococcus mutans obtained from the laboratory of Microbiology, Faculty of Medicine, Brawijaya University, Malang. The extract concentration used were 0%, 1%, 2%, 3%, 4%, and 5% v/v with four repetition. The concentration of Streptococcus mutans was 106CFU/ml. The statistic analysis showed insignificant differentiation between ethanolic extract of rome beauty apple peel and washington apple peel (Two Way Anova, p>0,05), yet there were showed significant effect of various concentrations of both extract (Post Hoc Tukey, p<0,05). MIC of both ethanolic extract of rome beauty apple peel and washington apple peel against Streptococcus mutans was 3% and for the MBC of ethanolic extract of rome beauty apple peel was 4% and washington apple peel was 5%. In conclusion, there is differences of antibacterial effectivity between ethanolic extract of rome beauty apple peel and washington apple peel extract towards Streptococcus mutans in vitro but it is not significant.
