Uji Ekstrak Etanol Daun Kemangi (Ocimum Sanctum L.) Sebagai Antibakteri Terhadap Streptococcus Mutans Secara In Vitro

Abstract

Karies gigi merupakan suatu penyakit jaringan keras gigi, yaitu email, dentin dan sementum serta memiliki etiologi multifaktorial seperti diet karbohidrat, mikroorganisme, host dan waktu. Beberapa mikroorganisme yang dapat menyebabkan lesi karies telah ditemukan, salah satunya yang paling dominan adalah Streptococcus mutans. Daun kemangi mengandung zat aktif yang dapat berfungsi sebagai antibakteri berupa eugenol, tannin, flavonoida, dan saponin. Tujuan penelitian ini adalah untuk mengetahui efek ekstrak etanol daun kemangi sebagai antibakteri terhadap Streptococcus mutans secara in vitro. Penelitian ini merupakan eksperimental laboratorik dengan metode difusi agar sumuran untuk mendapatkan zona hambat pada konsentrasi 12,5%, 25%, 50%, dan 100%. Hasil penelitian menunjukkan bahwa ekstrak etanol daun kemangi pada konsentrasi 12,5% mampu menghambat pertumbuhan Streptococcus mutans dengan rata-rata diameter zona hambat sebesar ±11,2325 mm, konsentrasi 25% sebesar ±14,4375, konsentrasi 50% sebesar ±17,165, konsentrasi 100% sebesar ±19,8675. Analisis data menggunakan uji One-Way ANOVA menunjukkan terdapat pengaruh signifikan dari pemberian berbagai konsentrasi ekstrak daun kemangi terhadap zona hambat bakteri Streptococcus mutans (p < 0,05). Uji Korelasi Pearson menunjukkan adanya hubungan yang kuat dan berbanding lurus antara perlakuan konsentrasi ekstrak daun kemangi (Ocimum sanctum L.) dengan zona hambat Streptococcus mutans (r = 0,812). Dari penelitian ini didapat kesimpulan bahwa ekstrak etanol daun kemangi (Ocimum sanctum L.) efektif sebagai antibakteri terhadap Streptococcus mutans secara in vitro.

English Abstract

Caries as a disease which affect hard structure of the teeth, called email, dentin and sementum and has multifactorial etiology such as carbohydrate diets, microorganisms, host and time. Several microorganisms that have cause lesion caries have been found, one of the most dominant is Streptococcus mutan. Ocimum sanctum L. Leaf contained an active substances that have function is antibacterial such as eugenol, tannin, flavonoida, and saponin. The target of the research is to determine the effect of Ocimum sanctum L. Leaf against Streptococcus mutans In Vitro. This research is laboratory experiment use diffusion agar well method, in order to make holes to get resistor zone is 12,5%, 25%, 50%, and 100%. The result of the research indicated that Ocimum sanctum L. Leaf extract at concentration 12,5% afford to inhibit the growth Streptococcus mutans with average inhibition zone diameter are ±11,2325 mm, concentration 25% average inhibition zone diameter are ±14,4375, concentration 50% average inhibition zone diameter are ±17,165, concentration 100% average inhibition zone diameter are ±19,8675. The data analysis used One-Way ANOVA, it is demonstrate that there are significant effect from applied concentration of Ocimum sanctum L. Leaf extract against inhibition zone Streptococcus mutans bacterial (p < 0,05). Pearson correlation test exhibit that there are strong relation and directly proportional to the concentration of Ocimum sanctum L. Leaf extract with inhibition zone of Streptococcus mutans (r = 0,812). In this research be obtained the conclusion that etanol extract Ocimum sanctum L. Leaf can be effective as antibacterial Streptococcus mutans In Vitro.
