Abstract

Escherichia coli merupakan salah satu mikroorganisme yang pada umumnya sering menyebabkan infeksi klinis pada saluran kemih, saluran empedu dan tempat lain di rongga perut. Maraknya infeksi yang disebabkan oleh bakteri ini menyebabkan banyaknya penggunaan antibiotik sebagai tatalaksana terhadap bakteri ini sehingga timbul resistensi terhadap antibiotik golongan tertentu. ESBL merupakan beta laktamase yang di mediasi oleh plasmid yang banyak ditemukan pada Escherichia coli. Kayu manis (Cinnamomum burmanii) dapat dipertimbangkan sebagai obat alternatif dari bahan alami karena mengandung flavonoid dan tannin yang memiliki efek antibakteri. Tujuan penelitian ini adalah untuk mengetahui apakah ekstrak Kayu Manis (Cinnamomum burmanii) bersifat antibakteri terhadap Escherichia coli ESBL secara in vitro. Penelitian ini merupakan eksperimental laboratoris dengan metode dilusi tabung untuk mendapatkan kadar hambat minimum dan kadar bunuh minimum pertumbuhan bakteri Escherichia coli ESBL. Konsentrasi ekstrak Kayu Manis (Cinnamomum burmanii) yang digunakan adalah 5%, 7.5%, 10%, 12.5%, 15%, 17.5%. Konsentrasi 0% atau aquades digunakan sebagai kontrol bakteri. Tiap perlakuan dilakukan pengulangan sebanyak 4 kali. Kadar hambat minimum didapatkan dengan mengamati kekeruhan tabung dan kadar bunuh minimum didapatkan dari perhitungan bakteri menggunakan colony counter. Analisis data menggunakan Uji Kruskal-Wallis dan Uji Mann-Whitney menunjukkan adanya perbedaan yang signifikan pada perubahan konsentrasi esktrak Kayu Manis (Cinnamomum burmanii) terhadap pertumbuhan bakteri Escherichia coli ESBL (p<0.05). Uji Korelasi Spearman menunjukkan adanya hubungan kuat dengan arah negatif yang dapat diartikan semakin meningkatnya konsentrasi esktrak maka pertumbuhan bakteri semakin berkurang. Berdasarkan penelitian ini, dapat disimpulkan bahwa esktrak Kayu Manis (Cinnamomum burmanii) memliki efek antibakteri terhadap Escherichia coli ESBL secara in vitro.

English Abstract

Escherichia coli is one of major microorganism generally caused clinical infections of the urinary tract, biliary tract and other sites of the abdominal cavity. The increase of infections caused by E. coli also raises the widespread use of antibiotics that further could lead to antibiotic resistance to certain groups. ESBL is a plasmid mediated beta-lactamase usually found in Escherichia coli. Cinnamon (Cinnamomum burmanii) can be considered as an alternative medicine from natural sources as they contain flavonoids and tannins that have an antibacterial properties. The purpose of this study is to determine whether Cinnamon (Cinnamomum burmanii) extract has an antibacterial effect for ESBL Escherichia coli in vitro. An experimental research is carried out using tube dilution method to obtain minimum bacteriostatic and bactericidal concentration of ESBL Escherichia coli. The Cinnamon (Cinnamomum burmanii) extract is varied at the concentration of 5%, 7.5%, 10%, 12.5%, 15%, and 17.5%. The concentration of 0% (aquadest) is used for control bacteria. Each concentration is applied with 4 repetitions. The minimum bacteriostatic levels obtained by observing turbidity of the tubes, while the minimum bactericidal levels obtained from the bacteria calculation using a colony counter. Data analysis using Kruskal-Wallis and Mann-Whitney tests show that there is a significant mean difference in the growth of the ESBL Escherichia coli(p <0.05). Spearman correlation test shows a strong correlation with negative direction, so the increasing concentration of the cinnamon extract declined the bacterial growth. Based on this study, it can be concluded that cinnamon (Cinnamomum burmanii) extract has antibacterial effect against ESBL Escherichia coli in vitro.
