Abstract
Disentri masih menjadi ancaman di Indonesia dan di seluruh dunia. Shigella menyebabkan shigellosis, atau disentri basiler yang menyebabkan diare pada usus, salah satu gejalanya berupa peningkatan sekresi cairan pada colon. Penelitian ini bertujuan untuk membandingkan berat colon mencit terligasi dan temuan histopatologi pada colon mencit yang dipapar S. dysenteriae dengan yang dipapar oleh S. sonnei. Sampel colon dibagi menjadi tiga kelompok, kelompok kontrol, kelompok yang dipapar S. dysenteriae, dan kelompok dipapar oleh S. sonnei. Penelitian ini dilakukan ex vivo menggunakan metode Mice Ligated Ileal Loop (MLIL). Variabel yang diukur adalah berat colon setiap 5 menit selama 30 menit yang mewakili penambahan sekresi cairan colon. Data dianalisis dengan One-way Anova, dan temuan histopatologi digunakan sebagai data tambahan untuk menunjukkan korelasi antara sekresi cairan colon dengan perubahan histopatologinya. Hasil penelitian menunjukkan bahwa terdapat perbedaan pada berat colon yang dipapar oleh Shigella dengan kelompok kontrol dalam waktu 30 menit dari pemaparan, namun secara statistik tidak signifikan. Temuan histopatologi tidak dapat digunakan sebagai data tambahan akibat kerusakan jaringan yang digunakan sebagai sampel menggunakan metode ini. Kesimpulan dari penelitian ini adalah terdapat perbedaan dalam berat colon yang dipapar oleh S. dysenteriae, S. sonnei, dan kelompok kontrol selama 30 menit dari pemaparan, namun secara statistik tidak signifikan. Dibutuhkan penelitian menggunakan metode lain untuk membandingkan gambaran histopatologi.
English Abstract
Dysentery still becomes a threat in Indonesia and worldwide. Shigella causes shigellosis, or bacillary dysentery that causes diarrhea in intestines, one of the symptoms is the increase of the secretion of fluid in colon. This study intends to compare the weight of ligated mice colon and histopathological findings of mice colon exposed to S. dysenteriae with ones exposed to S. sonnei. The colon samples were divided into three groups, the control group, group exposed to S. dysenteriae, and group exposed to S. sonnei. This research was done ex vivo using Mice Ligated Ileal Loop (MLIL) method. The variable measured is the weight of the colon every 5 minutes for 30 minutes which represent the addition of intestinal fluid secretion. Data were analyzed by One-way Anova, and histopathological findings are used as additional data to show the correlation between the secretion of colon fluid with its histopathological changes. The results showed that there were differences in weight between the colon exposed by Shigella and the control group within 30 minutes of exposure, but was not statistically significant. Histopathological findings can not be used as additional data due to damage of tissue used as the samples using this method. The conclusion of this study is there are differences in weight of colon exposed to S. dysenteriae, S. sonnei, and control groups for 30 minutes of exposure, but was not statistically significant. Another method is needed to compare the histopathological findings.
