Abstract

TGF-β adalah protein yang disekresikan untuk meregulasi proliferasi, diferensiasi dan kematian dari berbagai jenis sel, yaitu sel kekebalan, termasuk sel B, sel T dan sel dendritik serta makrofag mensekresi TGF-β. TGF-β dan PDGF adalah dua mediator yang paling penting untuk mengaktifkan HSC, sel HSC teraktifkan menjadi myofibroblas yang merupakan tipe sel utama menghasilkan ECM protein (kolagen tipe I, III dan IV). Produksi yang berlebihan dan deposisi protein ECM menyebabkan fibrosis hati. Kurkumin telah terbukti memiliki efek hepatoprotektif karena kandungan antioksidannya dapat menekan stres oksidatif serta menghambat perubahan HSC menjadi miofibroblas, sehingga dapat menekan produksi TGF-β. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian kurkumin terhadap penurunan kadar TGF-β jaringan hati pada tikus model fibrosis akibat paparan CCl4 pada minggu ke-2, 5, dan 9. Sampel adalah tikus Wistar jantan yang dibagi menjadi 8 kelompok, terdiri dari kontrol negatif (KN), kontrol positif (KP), kelompok perlakuan dengan lama pemberian kurkumin selama 2 minggu (KP2) dan kelompok kontrolnya (KK2), kelompok perlakuan dengan pemberian kurkumin selama 5 minggu (KP5) dan kelompok kontrolnya (KK5), kelompok perlakuan dengan pemberian kurkumin selama 9 minggu (KP9) dan kelompok kontrolnya (KK9). Dari hasil penelitian ini didapatkan rerata kadar TGF-β jaringan hati sebagai berikut KN (155,75 pg/mL), KP (276,58 pg/mL), KP-2 (90,83 pg/mL), KK-2 (155,17 pg/mL), KP-5 (214,75 pg/mL), KK-5 (274,17 pg/mL), KP-9 (209,42 pg/mL), KK-9 (272,75 pg/mL). Didapatkan juga perbedaan yang bermakna antara 8 kelompok perlakuan ini (one way ANOVA p = 0,000). Pada penelitian ini didapatkan penurunan kadar TGF-β jaringan hati pada pemberian kurkumin selama 2 minggu dan terjadi peningkatan pada pemberian selama 5 dan 9 minggu.

English Abstract

TGF-β is a secreted protein that regulate the proliferation, differentiation and death of various types of cells: immune cells, including B cells, T cells and dendritic cells, and macrophages secrete TGF-β regulating the proliferation, differentiation and activation by other cytokines. TGF-β and PDGF are two of the most important mediators to activate the HSC, HSC cells become activated to be myofibroblast, which is the major cell types produce ECM proteins (collagen type I, III and IV). Excessive production and deposition of ECM proteins may cause liver fibrosis. Curcumin has been proven having hepatoprotective effect because its antioxidants can reduce oxidative stress and inhibit the conversion of HSC into myofibroblast, so it can suppress the production of TGF-β. This study aimed to test the effect of curcumin therapy to TGF-β levels in liver tissue, using fibrosis rats model induced by carbon tetrachloride (CCl4) in week-2, 5, and 9 of curcumin administration. Samples are male Wistar rat which is divided into 8 groups that consist of negative control (KN), positive control (KP), the curcumin-given treatment for 2 weeks group (KP2) and it’s control (KK2), the curcumin-given treatment for 5 weeks group (KP5) and it’s control (KK5), and the curcumin-given treatment for 9 weeks group (KP9) and it’s control (KK-9). Results of this study, the mean levels of grup TGF-β on liver tissue showed KN (155.75 pg / mL), KP (276.58 pg / mL), KP-2 (90.83 pg / mL), KK-2 ( 155.17 pg / mL), KP-5 (214.75 pg / mL), KK-5 (274.17 pg / mL), KP-9 (209.42 pg / mL), KK-9 (272, 75 pg / mL). Significant differences found inall 8 grups (one way ANOVA p = 0.000). conclusion of this study is the levels of TGF-β decresed in 2 weeks curcumin treatment group. Inversely, increasing of TGF-β level found in 5 and 9 weeks curcumin treatment groups.
