Abstract
Lupus eritematosus sistemik merupakan kondisi inflamasi yang berhubungan dengan sistem immunologi yang dapat menyebabkan kerusakan multi organ. Insiden lupus meningkat hampir tiga kali lipat dalam 40 tahun terakhir. Infeksi masih menjadi penyebab penting dari angka mortalitas dan morbiditas pada pasien dengan LES dengan penyebab terbanyak adalah infeksi bakteri. Bakteremia sendiri merupakan penyebab penting dari morbiditas pada pasien LES. Saat ini sedang dikembangkan metode terapi baru yaitu menggunakan vaksin kinoid. Vaksin kinoid menekan aksi IL-17 yang merupakan sitokin proinflamasi sehingga keparahan penderita lupus berkurang. Tujuan dari penelitian ini adalah untuk membuktikan apakah vaksin kinoid IL-17A dapat menurunkan jumlah bakteremia pada mencit LES pasca injeksi MRSA (Methicillin-Resistant Staphylococcus aureus). Penelitian ini merupakan penelitian eksperimental yang menggunakan post test only group design dilakukan pada mencit Balb/c betina. Mencit diinjeksikan MRSA secara intraperitoneal dengan dosis 108 cfu/ml untuk dilihat bagaimana kerentanan infeksinya antara yang diberikan vaksin maupun tidak. Diperoleh data bahwa tidak ada perbedaan pengaruh yang signifikan pemberian vaksin kinoid IL-17A terhadap jumlah bakteremia pada mencit Balb/c model LES. Kesimpulan dari penelitian ini bahwa pemberian vaksin kinoid IL-17a tidak berpengaruh terhadap penurunan jumlah bakteremia pada mencit model LES.
English Abstract
Systemic Lupus Erythematosus is an inflammatory condition that is associated with the immune system that can cause multi organ damage. The incidence of lupus increased almost threefold in the last 40 years. The infection is still an important cause of mortality and morbidity in patients with SLE with the most cause is bacterial infection. Bacteremia is an important cause of morbidity in patients SLE. Currently being developed new therapeutic method using vaccine kinoid. Kinoid vaccine suppresses the action of IL-17 which proinflammation cytokine so the severity of lupus sufferers diminished. The purpose of this research is to prove whether vaccines kinoid IL-17A can decrease the amount of bacteremia on the SLE Balb/c mice post-injection of MRSA (Methicillin-Resistant Staphylococcus aureus). This research is experimental research using post test only group design done on female mice Balb/c. MRSA in murine injected on intraperitoneal with dosage 108 cfu/ml to know how the vulnerability of infection between given the vaccine or not. Retrieved data is that there is no difference significant influence the granting of kinoid IL-17A vaccines against bacteremia amount in SLE Balb/c mice. The conclusions of the research was that giving the vaccine kinoid IL-17A has no effect against a decrease bacteremia amount in SLE Balb/c mice.
