Abstract

Menurut Permenkes RI Nomor 722/Menkes/Per/IX/1988 tentang Bahan Tambahan Makanan, boraks merupakan salah satu bahan kimia yang dilarang digunakan untuk pangan. Namun, dalam penelitian yang dilakukan oleh Agus Pramono dalam Nasution (2009) tentang boraks pada beberapa jenis makanan, lebih dari 60% dari setiap jenis sampel positif mengandung boraks. Boraks merupakan zat aditif yang dapat melalui sawar plasenta dan dapat ditemukan di ASI, Apabila suatu zat asing dapat melalui sawar plasenta, zat tersebut dapat menjadi teratogen bagi janin. Menurut Cox (2004), asam borat sebagai hasil disosiasi dari boraks dapat menyebabkan berkurangnya jumlah ruas tulang belakang janin tikus, dan kegagalan pembentukan satu tulang rusuk. Sehingga, mungkin saja boraks dapat menghambat pertumbuhan tulang lain, contohnya tulang pada ekstremitas janin. Tujuan penelitian ini adalah untuk menguji apakah ada efek teratogen dari boraks terhadap morfologi ekstremitas bayi tikus Rattus norvegicus. Penelitian ini bersifat true experimental dengan rancangan Randomized Post Test Only with Control Group Design. Sampel yang digunakan adalah tikus wistar Rattus norvegicus yang berusia 8-10 minggu dan sehat. Sampel dibagi menjadi 5 kelompok dengan (5 ekor per kelompok), yaitu kelompok kontrol (tanpa paparan boraks), P1 (boraks sebanyak 50mg/kgBB), P2 (100mg/kgBB), P3 (200mg/kgBB), P4 (400mg/kgBB) sejak hari ke-4 hingga ke-17 kehamilan. Hasil: Dua ekor tikus kelompok perlakuan (masing-masing 1 ekor dari P1 dan P4) mati selama penelitian. 1 dari 209 fetus tikus lahir tanpa ekstremitas atas dan bawah (0,5%; p > 0,05); terdapat pemendekan panjang ekstremitas secara signifikan pada P1 (p < 0,05) dan peningkatan panjang ekstremitas secara signifikan dari kelompok P3 (p < 0,05). Jumlah fetus per induk dan kejadian kecacatan kelompok kontrol dan kelompok perlakuan tidak memiliki perbedaan secara signifikan (p > 0,05). Kesimpulan: pemberian boraks selama kehamilan dapat menyebabkan kecacatan, walaupun tidak secara signifikan.

English Abstract

As stated in Indonesian Health Ministry Regulation number 722/Menkes/Per/IX/1988 about Food Additives, borax is listed as one of several chemical compounds which is not allowed to use in food processing. But, the field survey done by Pramono in Nasution (2009) showed the fact that more than 60% of samples are borax-positive. As borax can surpass the placental barrier, it may become a teratogen for the growing fetus(-es). Cox (2004) mentioned that boric acid as a dissociated form of borax have effects in reducing vertebral segments and also caused the failure of ribs formation in rats. Since the affected sites are included in skeletal system, borax may have effect too in other regions of skeletal system, such as extremities. Objectives: this experiment was done to know any teratogenicity effects of borax on rat fetuses’ extremities morphology. This was a true experiment using Randomized Post Test Only with Control Group Design. The samples used were 25 rats which were divided into 5 groups: control group (without any borax administration), P1 (borax in amount of 50mg/kgW), P2 (100mg/kgW), P3 (200mg/kgW), and P4 (400mg/kgW). Borax solution in various doses were administered within the 4th – 17th day of pregnancy. Results: Two rats were died during the experiment, each from P1 and P4 group. 1 out of 209 fetuses was born without any extremities (0.5%; p > 0.05) and there was a significant shortening of extremities length in P1 group, and the vice versa occurred in P3 group (p < 0.05). Average number of delivered fetuses and the malformation occurrence were not affected by the borax dose (p > 0.05). Conclusion: exposures of borax during pregnancy may cause extremities malformation although this occurrence is not significantly happened.
